Cabbage white butterfly (Pieris rapae) is widely distributed in the whole north temperature zone,most areas in China, South India in South Asia. Due to the feeding habits and the various environment conditions, there are different geographical populations. By long-time isolation and evolution, there existed a certain degree of genetic difference between different cabbage white butterfly populations.The Amplified Fragment Length Polymorphism (AFLP) technique is used in this experiment to analysis the genetic diversity among five different geographic populations of Pieris rapae. We extracted the fragment of the original photograph and transformed them into a 0-1 matrix,then obtained the similar coefficient matrix. We analyze all individuals of different geographic populations of Pieris rapae by UPGMA, after standardizing 9 characteristic data of these 5 populations by SPSS15.0, then use the Between-group linkage method to analyze data. The results were as follows:We choose one primer combination to analysis the testing efficiency of silver nitrate staining and fluorescence marker method, the result showed that the fluorescence marker method can obtain 158 characteristic bands, with a polymorphic percentage of 94.49%, while using the silver nitrate method we obtained 67 characteristic bands, with a polymorphic percentage of 52.24%. We obtained 91 more characteristic bands from the fluorescence marker method compared with the silver nitrate staining, the testing efficiency of fluorescence marker method is 1.8 times to silver nitrate method.We got 540 bands altogether from the profile analysis,36 common bands and 504 polymorphic bands. The total polymorphic percentage is 93.33%. Different primer combinations can bring different amplified result, which means that the amplification efficiency is different.We analyze all individuals of different geographic populations of Pieris rapae by UPGMA, The result showed that:Taipingyu populations and Daxingshan populations was close and shared a cluster, some individual of Honghegu populations and Taipingyu populations and Daxingshan populatins were close, other individual of Honghegu populations were close to Niubeiliang populations, Niubeiliang populations and Xunyangba populations had a close relationship.Morphological data is not compeletely supportive to the results of molecular data, molecular data might be affected by geographic isolation, but morphology can be affected by the number of specimens, temperature, humidity and other factors.According to genetic differentiation analysis among 5 populations, which proposed that geographical isolation was locked by the Qingling Mountiains, different geographic populations had a relative high genetic differentiation.
