Objective:To explore the best duration time for the construction of tissue-engineering cartilage on the cyclic pressure.Methods:Free chondrocytes isolated from rabbit articular cartilage were seeded into PLGA scaffolds after expansion in vitro,and setted into 4 groups at random. Group 1~3 were cultured under cyclic pressure(0~200kPa, 0.1Hz)for 4hours﹑8hours﹑12hours;Group 4 was a control group(no pressure). Each group was cultured for 2 weeks.After 2 weeks of culture, the tissue engineering cartilages were observed in vitro and assessed by histological staining of H-E.Then,the content of DNA and the secretion of type II collagen and GAG in cartilages were detected in quantitative.Results:Under the cyclic pressure(0~200kPa, 0.1Hz),the group of 8hours is the biggest﹑lucidus﹑stretch and queuing discipline, II collagen and content of GAG also the highest (P<0.01). Conclusions:Chondrocyte is baro-senstive, the metabolism of chondrocytes is effected by the time of cyclic pressure, under the effect of 0~200kPa, 0.1Hz, cyclic pressure of 8 hours each day is better for cartilage bioplast multiplication and synthesis of extracellular matrix such as II collagen and GAG.
