Objective:To investigate the changes of gastric tissue COX-2 after intestinal perforations due to abdominal firearm wound. And to study its relationship with plasma endotoxin levels and pathological change of gastric tissue. Methods:A total of 42 Chang-Bai piglets were assigned randomly into 7 groups:control group and wounded 1,2,4,8,12,24 hours group.The model of intestinal perforations due to abdominal firearm wound was established in wounded groups.Gastric tissue COX-2 activity was measured with immunohistochemical s taining and image analys is in all groups.The plasma endotoxin levels were measured by chromogenic limulus amebocyte lysate test.The alterations of gastric tissue were observed under light microscope in all groups.Results:The expressions of COX-2 of gastric tissue in wounded groups were significantly increased compared with control group(P<0.05).Plasma endotoxin levels of the experimental group were significantly higher than that of control group (P<0.05),and the peak appeared from 8h to 12h after injury.The experimental group appeared gradually aggravated gas- tric mucosa hyperemia and edema under light microscope. Inflammatory cell infiltration and subs- erous hemorrhage appeared in 12h and 24h group. Conclusion: The expression of COX-2 in gastric tissue is increased while the change of gastric injury isaggravated with prolongation of wound after intestinal perforations due to abdominal firearm wound, which suggests that COX-2 may play an important role in gastric injury of pigs.
