OBJECTIVE To study the expression of hypoxia-inducible factor-1 alpha (HIF-1α),vascular endothelial growth factor(VEGF) and its receptor KDR,microvessel density(MVD) in endothelial carcinoma and investigate the impact of HIF-1α,VEGF,KDR and MVD, and the relation with HIF-1α,VEGF,KDR protein overexpression on the developmet and biological behavior of endometrial carcinoma.METHODS Expression of HIF-1 alpha,VEGF and KDR were analyzed by immunonistochemisitry and MVD was determined by CD34 immunostaininy in 39 cases of endometrial carcinoma,18 cases of endometrial hyperplasia and 17 cases of normal endometrium.And compared with clinical characteristics,such as age,clinical-stage and pathological grade et al.RESULTS HIF-1 alpha was overexpressed in endometrial carcinoma. Positive expression of HIF-1alpha was higher in endometrial carcinoma than that in endometrial hyperplasia and normal endometrium(x2=7.8222, P=0.0052;x2=24.777, P=0.0000). And Positive expression of HIF-1 alpha in endometrial hyperplasia was higher than that in normal endometrium (P=0.0408). Positive expression of VEGF in endometrial carcinoma was higher than that in endometrial hyperplasia and normal endometriam (x2=6.5183, P=0.0107;x2=16.1345, P=0.0001), and the expression of KDR in endometrial carcinoma was higher than that in endometrial hyperplasia and normal endometriam (x2=7.5693, P=0.0059, x2=17.1532, P=0.0000). MVD of endometrial cancinoma(46.77±10.67) is higher than that of endometrial hyperplasia(27.95±3.67) (t=7.2537, P=0.0000) and normal endometrium(17.04±5.35) (t=7.0705,P=0.0000).CONCLUSIONS The expression of HIF-1α,VEGF,KDR and MVD were increasing from normal endometrium,endometrial hyperplasia through endometrial carcinoma. In total, the results clue on the important role of HIF-1α,VEGF,KDR and MVD on the development of endometrial carcinoma.Additional we can treat them as endometrial carcinoma potential invasive tumor markers and as the anti-tumor therapy of important novel targets.
