Objective We examined cytochrome P450 2A13 protein and gene expression in oral cancer and their corresponding adjacent tissues,then analyzed the relationship between CYP2A13 expression and relevant clinical and pathologic data.Methods CYP2A13 mRNA was detected by real-time fluorescent quantitative PCR and Western blotting in 26 cases of oral cancer and their corresponding adjacent tissues. Using CEQ8000 sequencing instrument to identify the specificity of real-time quantitative PCR products. SPSS(14.0)statistical software was applied for data analysis. The correlation between the expression of CYP2A13 and the major clinical pathological characteristics of oral cancer were analyzed by rank test.Result 1.Real-time fluorescence quantitative PCR results: (1)The expression of CYP2A13 mRNA in oral cancer 22.1731 was significantly lower than their corresponding adjacent tissues 30.8269 (P=0.0202). (2)The expression of CYP2A13 in oral cancer:①male was 12.3846 and female was 14.6154(P=0.2397),②the patients under the age of 55 was 9.6, and those over the age of 55 was 15.9375 (P= 0.0204),③high histological grades was13.9524, middle and low histological grades was 11.6(P=0.2847),⑥lymphnode metastasis was 11.7692 and in no lymphnode metastasis was 15.2308(P=0.1321),⑦TNM stages(Ⅰ+Ⅱ)was 17.375 and TNM stages(Ⅲ+Ⅳ)was 11.7778(P=0.0452).Conclusion Down-regulated expression of CYP2A13 maybe involved in the development and progression of oral cancer due to the increased activity of certain carcinogens and the exact mechanism need further study.
