Purpose:The purpose of the study was to determine the maximal tolerated dose(MTD) and the dose-limiting toxicity(DLT)of Docetaxel combined with Capecitabine and concurrent radiotherapy as adjuvant treatment in patients with locally advanced gastric cancer.Methods and materrials:A total of 21 patients with advanced gastric adenocarcinoma after curative surgery were treated with radiotherapy to a total dose of 45Gy in 5 weeks. Docetaxel was administered at a dosage of 20mg/m2(n=3), 25mg/m2(n=3),30mg/m2(n=3),35mg/m2(n=6), and 40mg/m2(n=6), once a week.Capecitabine was administered at a dosage of 10000mg/m2,oral administration in twice daily when accept radiotherapy. DLT was defined as GradeⅢorⅣhematologic and nonhematologic toxicity.Results:GradeⅠ-Ⅲleukopenia, diarrhea, and nausea/vomiting were the most common toxic side effects, and most were Grade 1-2. A DLT was first observed in 2 of 3 patients at 35 mg/m2(1 of GradeⅢdiarrhea and 1 of GradeⅢleukopenia),and then increase 3 patients in the group, but was not observed in the same group.Then enter next 6 patients 40mg/m2 group.At 40 mg/m2, DLT occurred in 4 of 6 patients(2 of GradeⅢleukopenia and 1 of GradeⅢnausea/vomiting and 1 of GradeⅢdiarrhea). Toxicities included myelosupression, nausea, vomiting and hand-foot syndrome.Conclusions:Diarrhea were the most common dose-limiting toxicity (DLT). The maximal tolerated dose (MTD) of Docetaxel in this setting was 35mg/m2, ivgtt,once a week.
