Objective To study the influence of EGCG on the expression of NF-κB, MnSOD, Caspase-3 and its relationship with proliferation inhibition and apoptosis induction in nasopharyngeal carcinoma cell line CNE-2. This research was aimly to obtain a Sensitizing drugs combining with radiation to increase the therapeutic effect of NPC.Methods Nasopharyngeal carcinoma cell line CNE2 were cultured in vitro and treated with various concentrations of EGCG, the morphological changes of cells were observed by inverted microscope; the proliferation ability was detected by MTT; the apoptosis was observed by Hochest staining with the apoptosis rate counted; Flow Cytometry was used to analyze the changes of cell cycle; the expression of NF-κB,MnSOD,Caspase-3 were detected by one step Reverse Transcription Polymerase Chain Response(RT-PCR).Results 1. After treated with EGCG, The CNE2 cells became roll and small.increasing the concentration of the EGCG, some floating and dead cells can be seen in the inverted microscopy. EGCG could apparently inhibit the proliferation of CNE2 in a dose-and time-dependent manner(p<0.01). With the increasing of the concentration of EGCG and elongation of the treated time, The killing effect of EGCG become increasingly evident. 2. Apoptotic cells can be observe under the fluorescent microscope and the CNE-2 apoptosis ratio of control and EGCG treatment groups(40,80,160mg/l) were (2.3±0.7)%,(13.7±1.1)%,(22.5±1.2)%,(36.2±2.1)%,respectively. Compared to control group,the experimental groups can significantly induce apoptosis(p<0.01),the difference between experimental groups are also significant (p<0.01); 3. After treatment with various concentration of EGCG for 48h, the ratio of G1 phase cells of control and EGCG treatment groups(40,80,160mg/l) were (53.2±1.4)%,(64.8±1.2)%,(71.9±1.0)%,(74.5±1.6)%, respectively. Compared to control group,the experimental groups can significantly arrest the cell in G1 phase(p<0.01), the difference between experimental groups are also significant(p<0.01); 4.EGCG could down-regulate the expression of NF-κB,and up-regulate the expression of caspase-3 and MnSOD in a dose-dependent mannerConclusions EGCG can arrest the cells in G1 phase and induce apoptsis of CNE2 cells, the possible mechanisms were decreasing the expression of NF-κB, and enhancing the expression of MnSOD and Caspase-3...
