Objective To establish cell model ofβ2 -adrenoceptor downregulation in murine airway smooth muscle induced by salbutamol. So, on the basis of this platform, the molecular and biological mechanism ofβ2 AR downregulation will be elucidated in detail.Methods BALB/c mice ASMC were primarily cultured. Second to five generations of cells were identified by cell morphology and indirect immunofluorescence. They were divided into two groups at random: control group and salbutamool-treated group, which purity were more than 95% from third to fourth generations. The level ofβ2 AR mRNA and protein expression of control group and salbutamol-treated group were detected by RT-PCR and western blot.Results Primarily cultured cells were demonstrated typical characteristic of "peak and valley"-like growth trend. There were positive expression of specific smooth muscleα-actin filament paralleling to cell longitudinal axis in the cytoplasm. Their origin of smooth muscle cell had been validated which was consistent with their morphology and biological characteristics. The expression level ofβ2 AR mRNA in salbutamol-treated group was lower than that in control group ( P < 0.05), The expression level ofβ2 AR protein was also markedly lower than that in control group ( P < 0.05).Conclusions A cell model in airway smooth muscle cell related toβ2 AR downregulation had been successfully established. To lay a good foundation for furth study the mechanism ofβ2 AR downregulation in asthmatic patients.
