Objective: By collecting clinical specimens of normal bile duct and cholangiocarcinoma in patients, observed the expression of receptor for hyaluronan-mediated motility (RHAMM). Then discussed the role of RHAMM in invasion and metastasis of cholangiocarcinoma.Methods: According to the TNM staging system proposed by International Union Against Cancer (UICC): 0-Ⅱstage, no lymph node and distant metastasis.Ⅲ-Ⅳstage, invading adjacent tissue, or with lymph node metastasis, or with distant metastasis. We divided the speciments into three groups:○1 Normal Control group: Collected 15 cases of normal bile duct.○2 Cholangiocarcinoma group: Collected the tissue of cholangiocarcinoma in TNM staging 0-Ⅱ15 cases.○3Cholangiocarcinoma with metastasis group: Collected the tissue of cholangiocarcinoma in TNM stagingⅢ—Ⅳ15 cases. Then observed normal bile duct and cholangiocarcinoma tissue by pathology and used Immunohistochemistry and Western blot to assay the expression of RHAMM in each group.Results:○1 Immunohistochemistry: Positive expression of RHAMM were in Cholangiocarcinoma group and Cholangiocarcinoma with metastasis group with stained brown in the cytoplasm or membrane. RHAMM protein stained deepest in cholangiocarcinoma with metastasis group, followed by bile duct cancer group and was hardly seen in the normal control group.②Western blot analysis of the protein of RHAMM: The gray scale of RHAMM standardized by reference. The expression level of RHAMM was lowest in the control group (0.033±0.003), while increased in the cholangiocacinoma group (0.162±0.013). There were no significant difference between the two groups (P>0.05). The cholangiocarcinoma with metastasis group hold significantly higher expression than the other two groups, and the difference had statistical significance (P<0.05).Conclusion: All of above showed that RHAMM was not the decisive factor for change of normal bile duct to cholangiocacinoma. Because the significant increase in the group of cholangiocarcinoma with metastasis and a large number of them bind with its ligand HA, promoting the invasion and metastasis of cholangiocarcinoma.
