Objective To investigate the effect of Curcumin in acute lung injury (ALI) associated with severe acute pancreatitis (SAP) and explore the possible mechanisms.Methods 45 Sprague-Dawley (SD) rats were randomly divided into three groups: group A (shame operation group), group B (SAP group) and group C (SAP+ Curcumin group) each with 15 in group. Model was induced by modified method: SAP with ALI were induced by injection of 5% sodium deoxycholate through retrogradely common biliopancreatic ducts via papilla duodeni with epidural catheter, closed puncture hole with medical sealant glue. Rats were anatomized after 6 hours of injection. Visual observation of the general abdominal cavity and pancreatic pathology. Detection of arterial blood oxygen pressure (PaO2), carbon dioxide partial pressure (PaCO2) and serum amylase activity. Collection of pancreatic tissue, sliced observation. Extend the incision into the chest, visual observation of the broad chest and lung pathology. Take the left lung, measured lung wet / dry weight ratio. Take the right lung, making observation of paraffin sections: hematoxylin - eosin (HE) staining, observed under the microscope and score; SABC immunohistochemical detection of nuclear factor-κB (nuclear factor kappa binding, NF-κB) protein expression and semi-quantitative analysis. Results1 6 hours after surgery. B, C group pancreatic gross and microscopic changes in SAP were observed, and C lesions were compared with B group reduced.2 6 hours after surgery. B, C group lung gross and microscopic changes observed were ALI, and C lesions were compared with B group reduced.3 Three groups of microscopic pathology in lung tissue scores were: A group: 0.33±0.49, B group: 7.33±0.98, C group: 5.27±0.80; which A group <B group, C group (P <0.01) , B group> C group (P <0.01).4 Three groups of serum amylase activity were: A group: 1063.20±112.39 U / L, B group: 7319.27±201.37 U / L, C Group: 4252.87±199.41 U / L; where A group <B group, C group (P <0.01), B group> C group (P <0.01).5 Three groups of PaO2 were: A group: 95.67±2.97 mmHg, B group: 54.47±3.34 mmHg, C group: 73.47±4.03 mmHg; which A group> B group, C group (P <0.01), B group <C group (P <0.01).6 Three groups of PaCO2 were: A group: 37.40±2.90 mmHg, B group: 54.73±3.24 mmHg, C group: 45.00±2.48 mmHg; which A group <B group, C group (P <0.01), B group> C group (P <0.01).7 Three groups of lung wet / dry weight ratios were: A group: 4.13±0.34, B group: 9.05±0.39, C group: 6.14±0.28 mmHg; which A group <B group, C group (P <0.01), B group> C group (P <0.01).8 Three groups of lung tissue NF-κB p65 positive expression rates were: A group: 0%, B group: 100%, C group: 60%; which A group <B group (χ2 = 5.96 P <0.01), A group < C group (χ2 = 4.92 P <0.01), B group> C group (χ2 = 2.07 P <0.05).9 6 hours after surgery. expression of NF-κB of B, C group in lung tissue and the severity of lung injury was a significant positive correlation between (B group: rs = 0.589 P <0.01; C Group: rs = 0.803 P <0.01). Conclusion1 Acute lung injury in rats with severe acute pancreatitis, NF-κB in lung tissue over-expression, overexpression of NF-κB and the severity of lung lesions was positively correlated. Showed that NF-κB play an important role in the process of acute pancreatitis associated acute lung injury.2 Effect of Curcumin on severe acute pancreatitis associated lung injury has a protective effect, which play a role by inhibiting expression of NF-κB in the lung tissue.
