Objective:To study changes in efavirenz concentrations in whole blood and pharmacokinetics in rabbits before and after administration of ketamine. Methods:Twelve rabbits were first single administered with efavirenz, concentration of efavirenz in whole blood in different time were determined by liquid chromatographic-tandem mass spectrometric (LC-MS/MS). Ketamine had been administered, efavirenz were administered, efavirenz were determined by the above method. Results The diposition of efavirenz was conformed to two compartment model, after conbination with ketamine, efavirenz concentrations in whole blood all decreased, Cmax decreased(P< 0.05), t1/2 decreased(P< 0.05), AUC decreased(P< 0.05), C1 increased (P< 0.05) Conclusion:This paper by liquid chromatography tandem mass spectrometry results of stable and accurate, its specificity and precision are good. Ketamine decreases efavirenz concentrations in whole blood. Ketamine significantly accelerated the clearance rate. The combination of Ketamine with Efavirenz could induce pharmacokinetic interactions to reduce its anti-HIV effects.
