Purpose:To research the peak-expression time of BMP2,BMP7,TGF-β,bFGF,IGF-I, we detected the expression of them in different period of bone deficiency in test group and control group.And we can understand what is the relationship between these factors.Method:56 female Wistar rats were divided into test group and control group random, we took off both sides of ovary in test group,we just sew up the matched control group after open them, after 3 months, we duplicate osteoporosis model successfully.Do bone deficiency operation in both groups,extract the total RNA from 5 days,15 days and 30 days bone tissue,utilized fluorescence real-time quantitation PCR to detect the expression of BMP2,BMP7,TGF-β,bFGF and IGF-I. Bone tissue were taken to do paraffin section and HE stain, and then observe the morphological cells.Results:We discovered that the expression of each gene was different at each time stage in both test group and the matched group, the expression had statistical significance.In matched group,the expression peak-time of BMP2 was the 5 days.and the lowest expression was the 15 days.And in the test group,the peak-time was 5 days,the lowest time was 30 days. The expression had statistical significance in 5 days,15 days and 30 days (P<0.05).In matched group,the expression peak-time of BMP7 was 15 days;and in the test group,the peak-time was 5 days.the lowest time was 30 days. The expression had statistical significance in each time stage (P< 0.05). In matched group,the expression peak-time of TGF-βwas the 15 days,and the lowest time was 5 days.In test group,the peak-time was 15 days,.The expression had statistical significance in 15 days and 30 days (P<0.05).The expression peak-time of bFGF in both groups was 15 days,the lowest time was 5 days. The expression had statistical significance in each time stage (P<0.05) The expression peak-time of IGF-I in both groups was 15 days,the lowest time was 5 days. The expression had statistical significance in each time stage (P<0.05).In the observation of HE stain,the trabeculae architecture in matched group was a little changing.In the contrast to the matched control group, the test group was mess in trabeculae architecture, the thickness was thinner and the quantity was reduced, trabeculae was perforated,and had more vacant space, thinner cortical substance.Conclusion:The expression peak-time of most factors was 15 days after bone deficiency,but the BMP2 and BMP7 peak-time was 5 days which hinted that these two factors mobilized other factors,they were initiators. TGF-β,bFGF and IGF-I promoted the quantity of differentiation osteoblast,osteocyte and chondrocyte cells. They adjusted the differentiation and quantity of bone cells.The expression of bone deficiency factors in test group were lower than matched control group which hinted that it may took more time to repaire bone deficiency.
