Objective: To study the association between NRAMPl 3'UTR,FokI and TaqI of the VDR gene polymorphisms and susceptibility to tuberculosis of the Han of Xinjiang, and analysis the interaction between NRAMP1 gene and VDR gene.Methods: Using case-control study, we chased 216 patients with active pulmonary tuberculosis from the center for Disease Prevention and control of Tacheng. A total of 215 healthy people of the Han who have been sensitized to tuberculosis were selected as controls. Genotyping of the 3'UTR of NRAMP1 gene,FokI and TaqI of VDR gene was done by PCR-RFLP. Using Chi-square test to analysis the association between the three sites and tuberculosis,the calculations and statistics were performed with SPSS11.0.Results: 1. The frequencies of NRAMP1-3'UTR TGTG+/+,TGTG+/-,TGTG-/- genotypes in the case were 148 case (68.8%),52 case (24.1%),15 case (6.9%),which were 150 case (69.4%),36 case (16.6%),30 case (13.8%)in the control,the difference between the case and the control group had statistically significance(P<0.05).TGTG+/del+ genotypes had assignificant increased risk of tuberculosis.2.The frequencies of VDR-FF,VDR-Ff,VDR-ff genotypes in the case were 65case(30.2%),99case(46.1%),51case(23.7%)which were 114case(52.8%),86case(39.8%),16case(7.4%)in the control,the difference between the case and the control group had statistically significance(P<0.05).The frequencies of VDR-FokI-ff genotypes had statistically significant difference between the case and the control group(P<0.05); The frequencies of VDR-TT,VDR-Tt,VDR-tt genotypes in the case were 182 case (85%),26 case (12%),7case (3%),which were 188 case (87%),23 case (11%),5 case (2%)in the control,the difference between the case and the control group hadnot statistically significance(P>0.05). 3.There is positive interaction between NRAMP1-3'UTR and VDR-FokI; There is positive interaction between NRAMP1-3'UTR- GTGT+/-/ GTGT-/- and VDR-FokI- Ff/ ff(ORint=1.45)at the same time.Conclusions: 1. The NRAMP1-3'UTR allele may be a risk factor for the tuberculosis of han in Xinjiang; The han in Xinjiang carrying TGTG+/ TGTG- genotype are apt to suffer tuberculosis.2 .The FokI–ff polymorphism of VDR gene might be associated with TB in the Han of Xinjiang, The allele frequencies of may be a risk factor for the tuberculosis of the Han in Xinjiang. 3.But no evidences supported that the TaqI polymorphism were associated with TB in the Han of Xinjiang. The two polymorphisms didn't show linkage disequilibrium. 4. The interaction NRAMP1 and VDR gene may increase tuberculosis incidence of the Han in Xinjiang.
