Objective:To observe the Efects of DHL/RADA-16 Self-Assembling Peptide Nanofiber Scaffold on nucleus pulposus cells (NPCs) from rabbits.Methods:The NPCs were isolated from the immature nucleus pulposus (NP) of 2 Japan rabbits (3-week-old). The expressions of proteoglycan and collagen II of the NPCs were examined by toluidine blue and immunocytochemistry staining. DHL/RADA-16 was Self-Assembled by DMEM-F12. The NPCs were cultured with the Self-Assembling Peptide Nanofiber Scaffold. We observed the Effects of Self-Assembling Peptide Nanofiber Scaffold on NPCs. As a results the NP cells expressed proteoglycan and collagen II. Atomic Force Microscope indicated that the DHL/RADA-16 is the Self-Assembling Peptide Nanofiber Scaffold.The viability,proliferation and adhesion of NPCs were observed with inverted microscope, Calcein-AM / PI staining and Cell Counting Kit-8(CCK-8).Results: The NPCs cultured on the DHL/RADA-16 grew well and viable cell count was greater than 95%;The DHL/RADA-16 did no hinder the proliferation of NP cells and can promote the adhesion of NPCs.Conclusions:Self-Assembling Peptide Nanofiber Scaffold of DHL/RADA-16 has good biocompatibility and bioactivity for NPCs. It is a good biomaterial for NP tissue engineering.
