Objective:To detect DKK3 and SFRP2 gene methylation status between Uigur and Han ethnic groups and different breast disease (IDC, DCIS, ADH and UDH), we discussed the correlation Of DKK3 and SFRP2 gene methylation and breast cancer occurrence and development, different ethnic difference, and with the clinicopathological data.Methods:Using matrix-assisted laser desorption ionization time of flight mass spectrometry MALDI-TOF MS to detect DKK3 and SFRP2 methylation in Uighur breast samples 100 cases (IDC60 cases, DCIS 12 cases, ADH6 cases, UDH22 cases) and Han breast samples 100 cases (IDC50 cases, DCIS20 cases, ADH 10 cases UDH20 cases); data being proceed by SPSS 16.0 package GraphPad Prism 5 package for statistical analysis.Results:(1) DKK3 gene CPG4 sites methylation rate in IDC group and DCIS of the Uighur groups were higher than of the UDH group; (2) In the Uighur,SFRP2 gene CPG1.2 sites methylation rate in IDC group than of the UDH group. CPG3.4 sites methylation rate in the IDC group and DCIS group were higher than of the UDH group. CPG5 site methylation rate in the IDC group and DCIS group were higher than of the UDH group. CPG6.7 sites methylation rate in IDC group and DCIS group were higher than of the UDH group; In the Han, SFRP2 gene CPG1.2 sites methylation rates in DCIS and IDC group were higher than of the ADH group and the UDH group, CPG3.4 sites methylation rate in IDC Group was higher than of the ADH group. CPG5 sites methylation rate in IDC group is higher than of in ADH group and UDH group. CPG6.7 sites methylation rate in IDC group is higher than of in ADH group and UDH group, all data were statistically significant. (3) DKK3 methylation rate and rank analysis:IDC in the Uigur, CpGl site methylation rate was higher in≥45-year-old group than of in<45 age group, P value=0.0227 (p<0.05) is statistically significant; SFRP2 methylation rate rank analysis show that DKK3 gene CpG-1.2, CpG-3.4 and CpG-6.7 methylation rate were higher in of≥54 years of age group r than of<54 years group, and P values were less than 0.05, statistically significant. (4) SFRP2 gene all CpG sites methylation rate in IDC of Han is greater than of in Uighur, but only CpG 1.2 sites methylation rate was statistically significant (P <0.05).Conclusions:(1) The DKK3 gene CPG-4 site methylation of and SFRP2 gene CPG1.2, CPG3.4, CPG5 and CPG6.7 sites methylation may be early molecular event in abreast cancer. DKK3 and SFRP2 gene methylation may be involved in the development and progression of breast cancer. (2) SFRP2 gene CpGl.2 sites methylation rate in IDC group of Han were higher than of in Xinjiang Uigur, indicating possible differences with the national (3) DKK3 gene CPG1 site methylation in Uighur group IDC had significant differences of age,. SFRP2 gene CpG1.2, CpG3.4, CpG6.7 sites methylation had significant differences of age in the Han group IDC, suggesting that DKK3 and SFRP2 gene methylation and age may be relevant.
