Purposes: This subject study the cajal-like interstitial cells(ICCs) in guinea pig bladder. In this exper experiment We observe ICCs utrastructure and the relations with the surrounding nitric oxide positive neurons. We study the secreted functional characteristics of ICCs like cells by cell culture. In this subject can provide the function and significance of ICCs as bladder pacemaker cells. It can offer a new starting point linical and treatment of certain diseases (such as the detrusor instability, DI).Methods: In the experiment we use the immunofluorescence of c-kit to observe ICCs-like cells in normal adult guinea pig bladder tissue distribution. We make c-kit and NOS2 with immunofluorescence double-labeled technique to observe the relationship between ICCs-like cells and nitric oxide positive neurons . By enzyme digestion method and cell culture, we use c-kit and NOS2 immunofluorescence double staining technique to explore the secretory function of the ICCs-like cells.Results: In this experiment the ICCs can be observed much more in muscularis than submucosa and mucosal layer , as while as bei more in bladder top than bladder body and bladder neck. With the double-labeled we can observed ICCs-like cells distribute in the surrounding of the nitric oxide-positive neurons. With Immunofluorescence double staining of ICCs-like cells, we can find c-kit and NOS2 co-expressed in ICCs-like cells.Conclusions: The ICCs in guinea pig bladder are distributed in different parts in line with physiological functions, which lay the basis as the pacemaker cells. ICCs-like cells scatter in the surrounding positive neurons of nitric oxide, and they are both located in the bladder smooth muscles. This sturdy provides a strong evidence of"nerve - ICCs-like cells - smooth muscle". With Immunofluorescence double staining of ICCs-like cells, we find ICCs-like cells can produce NO. The experiment is very useful in the secretion function of ICCs-like cells. It also plays an important part in detrusor. It can offer a new starting point linical and treatment of certain diseases.
