Objectives:Traditionally, radiochemotherapy has been the choice of treatment for cervical carcinoma;however,it also produces serious side effects such as hemorrhage and infection. The side effects dramatically impact patients'quality of life and discourage long-term use of therapy. There is, consequently, a critical need for novel, more effective drugs with fewer side effects in cervical carcinoma therapy. Gossypol, a natural contraceptive drug with fewer side effects, has recently been proposed to be a potent drug to treat some neoplasms. To investigate the effect of gossypol and gossypol -based combination DDP on Hela cells of cervical carcinoma.Methods:The sensitivity test of gossypol,DDP and gossypol combination with DDP, was carried out by MTT assay to observe the cell growth inhibiting rate; Cell apoptosis was observed by fluorescence microscopy-acridine orange/ethidium bromide double staining and flow cytometry. Cell protein of Bcl-2 and Bcl-xL was observed by immunohistochemistry.Result:The result of MTT assay indicated gossypol was effective in inhibiting the proliferation of Hela cells. The increasing effect time and concentration of gossypol to result in the increasing inhibition rate. The inhibiting rates of DDP to Hela cells were enhanced significantly by the combination with gossypol. The result of AO/EB double staining and flow cytometry showed that gossypol and DDP alone or in combination induced Hela cells apoptosis significantly. The effect of inducing apoptosis of gossypol and DDP in combination was significantly more than them alone. The result of immunohistochemistry indicated that gossypol and DDP alone or in combination down-regulated expression of Bcl-2 and Bcl-xL. The effect of down-regulating expression to Bcl-2 and Bcl-xL DDP were enhanced significantly by the combination with gossypol.Conclusion:Gossypol effectively inhibits the Hela cells of cervical carcinoma and induces its apoptosis. The sensitivity can be enhanced by the combination of gossypol with DDP.
