Objective:To investigate the effect of transfection and expression of p27 gene on cultured vascular smooth muscle cells(VSMCs) in rats, and confirm the specificity driven by SM22alpha gene promoter.Methods:Recombinant lentivirus vector,Lenti-SM22alpha-p27,containing rat SM22alpha promoter and human p27 gene was constructed by site special recombination and was used to transfer p27 gene into rat VSMCs. After infecting cells for 48h, the exogenous p27 protein expression and its effects on cell proliferation were determined by BrdU and MTT assay.Results:The titers of resultant recombinant lentiviruses was 2.0×106TU/mL. p27 protein was over-expressed in VSMCs infected with Lenti-SM22alpha-p27, resulting in inhibition of cell proliferation. No p27 over-expression and inhibition of proliferation was det ected in non-vascular smooth muscle cells(NSMCs).Conclusion:The overexpression of p27 driven by SM22alpha can significantly inhibit the proliferation of rat VSMCs,without significantly effect on NSMCs.
