Objective targeted nano magnetic resonance contrast medium against epidermal growth factor receptor (EGFR) was prepared for in-vitro test. Methods etuximab (c225) and ultrasmall superparamagnetic iron oxides (USPIO) were connected by carbodiimide to form targeted nano contrast medium C225-USPIO against EGFR. Nasopharyngeal carcinoma cell line with high-expressed EGFR was selected and the immunobinding activity of C225-USPIO and CNE1 were detected by flow cytometry and indirect immunofluorescence. Targeted binding and ingestion statuses of CNE1 cell to C225-USPIO were detected by Prussian blue staining and transmission electron microscope (TEM). MRI signal strength variation was detected by 4.7T magnetic resonance imaging (MRI), after C225-USPIO with Fe concentration 50μg/ml incubating CNE1 cells for different time gradients.Result as indicated by flow cytometry and indirect immunofluorescence, C225-USPIO had well immunobinding activity with EGFR on the surface of CNE1 cells. And as indicated by Prussian blue staining as well as transmission electron microscope (TEM) detection, C225-USPIO could specifically bind with EGFR on the surface of CNE1 cell and be ingested into the cell. As indicated by 4.7T MRI, CNE1 could bind and ingest C225-USPIO, thus reducing the signal strength of MRI T2 significantly.Conclusion C225-USPIO could specifically bind with CNE1 cells with high-expressed EGFR, have well specificity and targeting and could reduce the T2 signal strength of magnetic resonance significantly, thus providing a sufficient experiment basis for in-vivo experiment.
