| Objective To research the effects and changes of expression of PSD95 (postsynaptic density-95) , Synapsin I and the phospho state of which in rat's gastric antrum with diabetic gastroparesis(DGP),in order to investigate plasticity of enteric nervous system (ENS).Methods 48 SD (Sprague Dawley) male rats were used in this study, randomly divided into diabetic group (n=32) which received intraperitoneal injection of STZ (streptozocin), and control group (n=12) with injection of citric acid buffer. Models were all sacrificed at the time point 2, 4, 8, 12 weeks after injected with STZ or citric acid buffer correspondently. Rat gastric antrums were obtained and then Real-time PCR and Western Blot were used to detect the expression of PSD95,Synapsin-I,P-Synapsin-I in diabetic rats at 2wk, 4wk, 8wk, 12wk respectively. RNA level and protein level were finally obtained with data analysis.Results In the diabetic model group,the RNA and protein expression of PSD95,Synapsin-I,P-Synapsin-I in DM groups were decreased compareing with that in the control group (P<0.05), and expression were decreased gradually as the disease progressed from 4wk to 12wk (P<0.05). Meanwhile, there was no significant difference detected in the expression of RNA or protein among control models at different time point (P>0.05).Conclusion One of the basic pathomechanisms of DGP is the disorder of ENS, therefore two important syanptic-associated proteins-PSD 95 and Synapsin-I were used for study. In the progression of DM, the expression of PSD95 and Synapsin-I are decreased, which may contribute great importantances to disfunction of ENS and progression of DGP. PSD 95 and Synapsin-I can also influent peripheral nervous system a lot, besides maintaining the synaptic function of central nervous system as well known. We can look forward to new findings in the treatment of DGP. |
