Objective: This paper is to observe the effect of Nur77 gene expression in hyperplastic vascular smooth muscle cells (VSMC) stimulated by platelet-derived growth factor (PDGF-BB), and to investigate whether the expression of Nur77 related to ERK signal transduction pathway.Methods: Primary rat aorta media smooth muscle cells were cultured in vitro use the method of adherence vascular tissue piece.The cells were grouped into three as control group, PDGF-BB group and PDGF-BB+PD98059group randomly. Protein level of Nur77, PCNA and ERK1/2 was investigated after the stimulus in 1h, 2h,4h and 8h with Western blot analysis .Results: The results showed that Nur77 protein level was related to VSMCs proliferation which stimulated by PDGF-BB. Western blot analysis shows that the ERK inhibitor PD98059 decrease the expression of Nur77 and PCNA protein level. Conclusions:There is increased expression of Nur77 protein during VSMC proliferation stimulated by PDGF-BB, and the expression of Nur77 is potentially mediated by ERK signal cascades.
