Objective To study the expression and activation of NF-ΚBp65 in peripheral blood mononuclear cells in patients with systemic lupus erythematosus(SLE) and their correlation with immunological index and glucocorticoid.Methods According to 1997 American College of Rheumatology SLE classification criteria,we would plan to choice Wuhan Union Hospital out-patients and in-patients who satisfied experimental standard. All cases were based on SLE disease activity index (SLEDAI) to 0 ~ 3 minutes as a non-event, 4 ~ 9 pm classified as mild activity, 10 to 14 minutes as a moderate activity, greater than 15 was severe activity . At the same time , we would set up a normal control group. Mononuclear cells were extracted from health adult and SLE patients'peripheral blood. NF-κBp65 expression and activation were detected by the method of immunocytochemical in different groups.Results①T here was expression of NF-κB in cytoplasm and nucleus on active SLE patiants'peripheral blood, but there was only little expression of NF-κB in cytoplasm on inactive SLE patiants and health adult peripheral blood.②NF-κBp65 expression and activation were positive correlation with the anti-dsDNA antibody and anti-Sm antibody positive rate, SLE disease classification, respectively, there were negative correlation with complement C3,C4 .③The expression of NF-κBp65 and activation were statistically significant in glucocorticoid-sensitive group and glucocorticoid-resistant groups.Conclusion The expression of NF-κBp65 and activation had a positive correlation with active SLE disease classification in SLE patients'peripheral blood; Glucocorticoid can reduce the amount of NF-κBp65 expression and activation.
