| Objective: To investigate can or should not rat bone mesenchymal stem cells(MSCs) that induced by rat,s serum contained chinese medicinal formulae qingnaoling differentate to neuron—like cells in vitro。Methods:Got the rat,s femoral bone in the condition of germ free,abscised brawn,periost,apophsis bone and run-out the bone marrow by LG-DMEM with 20% FCS into asepsis utensil. Rat bone mesenchymal stem cells were separated and got from rat bone marrow.Got the purified rat bone mesenchymal stem cells by whole blood wall sticking method and density gradient centrifugation(percoll,s method),amplified and cultured rat bone mesenchymal stem cells in vitro to get a number of rat bone mesenchymal stem cells. Test we experiment got rat,s bone mesenchymal stem cells can or should not express surface antigens heterophil granulocyte cell adhesion factor CD11b and lymphocyte factor CD45 used flow cytometry to accredit we have got rat,s bone mesenchymal stem cells is real or not. Rat bone mesenchymal stem cells were induced by rat,s serum contained chinese medicinal formulae qingnaoling that density was 1mg/ml. Observed the induced bone mesenchymal stem cells after 5 hour,12 hour,1day,3days,7days under phase contrastmicroscope.Induced bone mesenchymal stem cells were identified with nestin,glial fibrillary acidic protein (GFAP)and neuron-specific enolase (NSE) by immunohisto- chemistry. Results : The rat,s bone mesenchymal stem cells is unexpressed heterophil granulocyte cell adhesion factor CD11(b1.76%),lymphocyte factor CD45 (2.02%)by tested through flow cytometry. Immunolhistochemistry express neuron-like cells that is rat,s bone mesenchymal stem cells induced by rat,s serum contained chinese medicinal formulae qingnaoling nestin and NSE positive, GFAP negative。Conclusion:The rat,s bone mesenchymal stem cells can be induced to differentiate into neuron-like cells by rat,s serum contained chinese medicinal formulae qingnaoling in vitro. |
