OBJECTIVE: To investigate the influence of Neurogranin(Ng)on the calcium / calmodulin-dependent protein Kinase on the Ca2+ signal in peripheral blood mononuclear cells (peripheral blood nuclearcells, PBMCs) with SLE, we have detected the mRNA and the protein expression of Ng and CaMK2 and CaMK4.METHODS: Samples from 31 SLE patients and 18 healthy controls were collected. RT- PCR and Nested-Polymerase chain reaction (Nested-PCR) was used to examine the mRNA levels of Ng andα,β,γ,δCaMK2 and CaMK4, while Western blot analysis was used to examine the proteins level of CaMK2 and CaMK4.RESULTS: SLE patients exhibited a 90.3% expression of Ng mRNA , the control group exhibited a 55.5% expression. Ng-positive SLE patients 100% expressed CaMK4 mRNA, Ng negative samples 54.5% exhibited a expression of CaMK4 mRNA. 100% Ng positive SLE patients expressedγCaMK2 mRNA, 89.3% expressedδCaMK2 mRNA; control group 80.0% expressedγCaMK2 mRNA, 30.0% expressedδCaMK2 mRNA. Ng positive SLE patients 89.3% exhibited a co-expression ofγCaMK2 andδC CaMK2 mRNA, the expression of the control group were 30.0%.ONCLUSIONS: Neurogranin(Ng)was ectopic over-expressed in peripheral blood mononuclear cells in SLE patients. Ng-positive SLE patients highly expressed CaMK4 mRNA,γCaMK2 mRNA,γCaMK2,δCaMK2 mRNA.
