Gliomas are the most commen primary brain tumors. The extensive heterogeneity of the tumors has made their pathological classification rather difficult. Approaches for glioma diagnosis can be highly subjective due to the heterogeneity and infiltrative nature of these tumors and the dependence on the skill of the neuropathologist.We investigated differential expression proteins between low-grade astrocytomas and its periphery by proteome approach based on two-dimensional gel electrophoresis and matrix-assisted laser desorption/ionization time-of-flight /time-of-flight mass spectrometry. The stained gel images were analyzed with the PDQUEST software. 64 protein spots differentially expressed were found. These protein spots were digested with trypsin enzyme and then subjected to matrix-assisted laser desorption/ionization time-of-flight /time-of-flight mass spectrometry analysis. We identified 25 proteins, of which, 16 proteins were up-regulated and 9 proteins were down-regulated in low-grade astrocytomas compared to the periphery.Among these proteins, 6 of which are heat shock proteins, 7 metabolism related proteins, 3 brain proteins, and 10 proteins involved in other cellular functions. RT-PCR, western blot and immunohistochemical analysis were explored to confirm the differential expression of the transthyretin and glial fibrillary acidic protein. Transthyretin is a small molecular secret protein involving in transporting thyroxine from the bloodstream to the brain. It may be a candidate biomarker for the diagnosis of astrocytomas.ABSTRACT 2In search of mechanisms and function of TRIM45 gene, it was introduced into Hela cells. Differential display proteomics techniques was employed to study protein changes of Hela cells transfected with pCMV-Tag2B-TRIM45 compared with cells transfected with pCMV-Tag2B. Image analysis was carried out using PDQuest 7.2 image analysis software. 15 proteins were identified, of which 8 were up-regulated while 7 were down-regulated. These in vitro studies may be helpful in studying mechanisms that may lead to the function of TRIM45.
