Quercetin is the common flavonoid compound in dietary plan.It is widely distributed in various plants and have many biological activities,such as antioxidant, anti-mutagenic activities,and free radical scavenging function.In recent years,studies on Quercetin has become the hot spot.Molybdenum is one of the lanthanides and functions as the cofactor of some enzymes in vivo.Studies on Chinese herb medicine found that the purified herb medicine showed more side effects than the complex.Cao zhiquan,a professor in Beijing University created the coordinate theory in Chinese herb medicine in 1995.He pointed out that pharmacology activity of nature herb comes from the coordination of organic components and minerals of the herbs.The reactions and bio-activity between quercetin and several metal ions have been reported,but little is known about the reaction and bio-activity between quercetin and Mo(Ⅵ).The present study was carried out by using the single-factor method and multi-factor orthogonal experiment to find out the best condition of molybdenum synthesis,such as the molar ratio of the two substrates,reaction pH,temperature,time length.The quercetin-Mo(Ⅵ) complex was synthesized under the best condition.The synthesized products were analyzed by UV,IR spectrum.The coordination ratio, acute toxicity,genetic toxicity,superoxide anion(O2-·) and hydroxyl radical(·OH) anti-anion activity were also tested. Objectives1.To investigate the best reactive condition between quercentin and molybdenum,and try to find the possible structure of the product by using UV,IR spectrum measurement.2.To evaluate the toxicity of Quercetin-Mo(Ⅵ) complex by using the acute toxicity and micronuclei tests.3.To test the free redical scavenging activity of Quercetin-Mo(Ⅵ) complex.Methods1.The single-factor method and orthogonal experiment were used to search the best reactive pH,temperature,duration and the molar ratio of substrates Quercetin and Mo(Ⅵ) complex was synthesized at the best condition and its structure was estimated.2.The toxicity of Quercetin-Mo(Ⅵ) complex was evaluated by using the acute toxicity and micronuclei tests.3.The free radical scavenging effect of Quercetin-Mo(Ⅵ) complex was measured by using improved pyrogallic acid and ortho-oxybenzoic acid methods.4.Data were analyzed by using SPSS12.0 software.The mean of mice body weight and the rate of micro-nuclear were analyzed by t-test and X2 respectively. The significant level was set at a=0.05.Results1.The maximum yield of Quercetin-Mo(Ⅵ) complex was about 90%by using ethanol as the solvent,the reaction for six hours,the molar ratio of substrates from1.5 to 2.0(quercetin:ammonium molybdate),at pH 2.0 and 30℃.The possible structure of the complex was investigated by using IR,UV and fluorescence spectra methods.The apparent stability index was calculated by molar ratio and balanced moving methods,K=1.0×108.2.No poisoning signs were observed in the experimental mice when the complex dosage was introduced at 10.00g·kg-1.The positive control group showed higher bone marrow cell nuclear rate than the negative control group and experimental group (P<0.05).The experimental group and the negative control group of nuclear-rate was in the normal range.3.The Quercetin-Mo(Ⅵ) complex showed higher free radical scavenging effect for O2-·than Quercetin at low concentration,but lower at high concentration.At both high and low concentrations the Quercetin-Mo(Ⅵ) complex showed stronger effect for·OH scavenging.Conclusions1.The study established the best coordinated reactive condition of the synthesis of Quercetin-Mo(Ⅵ) complex,investigated the structure of the complex.Under the best condition,the highest yield of Quercetin-Mo(Ⅵ) complex are achieved.2.Under current condition,there were no signs of acute toxicity and genetic toxicity were found.The complex showed strong power of free radical scavenge.
