Radiotherapy is one of the important conventional method to treat the tumor.radiotherapy use the direct way to break the DNA by particle and photon,and also (or)use the indirect way which ionize water molecules to produce free radicals which play a curative effect to kill the tumor cells. The different ray sensitive cell lines (including adherent cells:MCF-7; HepG2; Hela; Glc-82; B16; SW-480, non-adherent cells were EL-4) was used in the present work.By western hybridization and immunoprecipitation method,we discussed the sumoylation of NOX family.we tried to reveal the different doses of X-ray and 12C heavy ion whether impact the expression and activity of NOX family of NADPH oxidase which is the key oxidase to produce ROS.we also discussed the empression and activity of NOX family of NADPH oxidase between different cell lines without any treatment.1. Different doses of X-ray radiated cells, NOX family of NADPH oxidase have performanced different radiation sensitivity. In addition to blood cells (EL-4),the expression of NOX1 and NOX4 unchanged.other cells NOX2, NOX3 and NOX5 of NADPH oxidase express in the different degrees of change.2.The NOX family of NADPH oxidase have different radiation sensitive between the mouse tumor cell (B16) and other adherent cells of human tumor cells after the X-ray radiation.3.The NOX family of NADPH oxidase have different radiation sensitive between the blood cell (EL-4) and other adherent cells of human tumor cells after the X-ray radiation.4. Tumor cells (HepG2, Hela, Glc-82) in the normal state,the expression level of the NOX family of NADPH oxidase are differences.5. Different doses of 12C heavy ion radiated cells(HepG2, Hela, Glc-82), NOX family of NADPH oxidase have performanced different radiation sensitivity. Radiation therapy combined with medication therapy, especially combined with heavy ion take more and more attention, Experiment provided the basis of the relationship between NOX family NADPH oxidase and the radiosensitivity of tumor cell, and then to provided a new way of gene therapy for sensitization to tumor cells or protection tonormal cells to protect.
