Transplantation of bone marrow mesenchymal stem cells with adrenomedullin (ADM) gene and hepatocyte growth factor (HGF) gene transfection was regard as one of the most promising techniques of treating AMI and CHF secondary to AMI. But the capcity of survival of these MSCs are impaired in absence of oxygen environment. And it is the key problem we must solve.Objective Compare of apoptosis of bone marrow mesenchymal stem cells with ADM gene and HGF gene transfection.Methods MSCs were incubated in the medium containing serum-free medium (control group), empty adenoviral vector (Ad-Null group), ADM gene modified adenoviral vector (Ad-ADM), HGF gene modified adenoviral vector (Ad-HGF) respectively. The levels of ADM, HGF, vascular endothelial growth factor(VEGF)of MSCs which were anaerobic cultivated for 0 hour, 3 hours, 6 hours, 9 hours, 12 hours, 16 hours, 20 hours and 24 hours were determined by ELISA respectively. The apoptosis index of MSCs which were anaerobic cultivated for 0 hour, 3 hours, 6 hours, 9 hours, 12 hours, 16 hours, 20 hours and 24 hours were measured by TUNEL.Results ADM and HGF gene transfection promoted the expression of ADM and HGF in MSCs. When all groups were anaerobic cultivated for 0, 3, 6, 9, 12, 16 hours, the level of apoptosis index of Ad-ADM group and Ad-HGF group were less than control group (P<0.05) and Ad-Null group (P<0.05). And When all groups were anaerobic cultivated for 9 hours, the level of apoptosis index of Ad-ADM group were began to less than Ad-HGF group (P<0.05). When all groups were anaerobic cultivated for 6 hours, the level of VEGF of Ad-ADM group and Ad-HGF group were began to higher than control group and Ad-Null group (P<0.05). When all groups were anaerobic cultivated for 9 hours, the level of VEGF of Ad-ADM group were began to higher than Ad-HGF group (P<0.05). The releasing of ADM and HGF were peaked at aerobic cultivation for 12 hours in Ad-ADM group and Ad-HGF group.Conclusions After transfection of ADM gene or HGF gene, the apoptosis of MSCs decreases. ADM gene transfection could reduce the apoptosis more effectively than HGF gene transfection which may be related to the expression of VEGF.
