Objective:To investigate the influence of ischemical reperfusion injury in intrahepatic bile duct on liver under liver transplantation.Methods:The rabbits were randomly divided into 5 groups:SO group, HBO1.5h group, HBO2h group, HBO2.5h group and HBO3h group. HBI was performed for Oh,1.5h,2h,2.5h and 3h respectively. Blood samples and tissue specimens were taken to determine the changes in liver function, energy metabolism and hepatobiliary histopathology before ischemia and after reperfusion for Oh,12h, 1d,2d,3d and 7d. The mortality was noted within the 7-day period of experiment. A 100% 7-day survival rate along with the functional and structural changes were taken into account to decide the tolerance limit, the maximum safe length of ischemiaResult:Hepatic cell and bile duct epithelial cell degree of injury gradually aggravate, appearance in liver function and postop 7 day death rate raise up, with time of ischemia temporal extension prolonged。The dominant reciprocity in injury liver and gall cellularoedema and phlegmasia cell infiltrate ect, within HBO2h group biliary tract I/R。The damage characteristic is biliary tract exuviation and necrosis focus of infection emergeirreversible in remain group above HBO2h group biliary tract I/R。Conclusions:The present study demonstrates the safety tolerance limit of biliary blood inflow occlusion induced by CBD and CHA clamping in rabbits is about 2 hours.
