Objective: To study the effect of lycopene in Tomato Oleoresin and grape seed extract to reduce the role of serum lipid profile and their unification, so as to give the basic datas for research and develop the health food. Methods: (1)Twenty male and ten female, twenty male wistar rats were divided into two groups , these two groups were the first and the second group in our experiment.Ten female wistar rats were the third group. The first group fed by lipid emulsion which included 10% cholesterol. The second and the third group fed by lipid emulsion which included 20% cholesterol .The serum TC, TG, HDL and LDL were tested at the zero day and fifthteenth day respectively.(2) Established a hyperlipemia rat model using lipid emulsion, according to the orthogonal design(L4(23)) and the serum TC, twenty male wistar rats were divided into 4 groups randomly, each group had 5 rats. On the basic of using lipid emulsion , the rats were given seed fat, 10mg·kg-1·d-1 of lycopene in Tomato Oleoresin, 100mg·kg-1·d-1 Proanthocyanidins in grape seed , the same dose of the mixture with lycopene in Tomato Oleoresin and grape seed extract separately. All groups were given reagent for 30 days, tested the serum LDL,HDL,TG,TC at the forty-sixth day.(3)One hundred wistar rats (200±20g)were divided into ten groups by weight randomly: normal control group, solvent blank group,high-lipid model group, lycopene in Tomato Oleoresin group, grape seed extract group, high-lipid model and lycopene in Tomato Oleoresin group, high-lipid model and grape seed extract group, low, medial and high dose group of high-lipid model and lycopene in Tomato Oleoresin and grape seed extract group. Normal control group, solvent blank group,lycopene in Tomato Oleoresin group, grape seed extract group used distilled water, other groups used high-lipid emulsion to establishe hyperlipidemia rat model. From the sixteenth day, on the basis of continue establish model, all groups were given reagent for 30 days, rats were killed after their blood and liver were taken out at the forty-sixth day, the serum LDL,HDL,TG,TC,MDA,NO content , SOD,GSH vitality were tested; weigh their liver , calculate liver coefficient. Result: (1): The rats fed by lipid emulsion which included 10% cholesterol at fourteen day, the serum TC and LDL were higher than the incipient level(P<0.05). The rats fed by lipid emulsion which included 20% cholesterol , the serum TC,TG and LDL of the female and male wistar rats were higher and with statiatical significance(P<0.05). (2) When the effect to decreace blood fat of lycopene in Tomato Oleoresin and grape seed proanthocyanidin extract was not obviously, their interaction to TC,LDL,HDL was obviously, the result was significantly difference. (3) Completely randome result showed us: compared with high-lipid model group, M+LP+GSE low dose group and M+LP+GSE media dose group serum TC,LDL decreaced(P<0.05), serum NO content and GSH vitality increaced(P<0.05). Conclusion: (1) Establishing a hyperlipidemia rat model successfully by using lipid emulsion,when the male rats were fed by lipid emulsion which included 20% cholesterol had obvious effects ,that were suitable for the reseach of mixed hyperlipoidemia,Hyperch- olesteremia,hypertriglyceridemia.(2)The combination of grape seed extract and lycopene in Tomato Oleoresin can reduce the serum lipid profile,and combined has a better effect.This effect is related with their antioxidation possibility.
