Objective: To investigate the mechanism of Astragalus Danshen compound on rat submandibular salivary glands in a model of Sjogren's syndrome (SS), and to take its expression of aquaporin-5 for target point.Methods: Submandibular gland tissue was extracted from Wistar rat before blending and emulsification with complete Freund's adjuvant (CFA), After that, subcutaneous injection of diphtheria-pertussis and tetanus vaccine (DPT) was conducted on the back to enhance immunity. Through perfusion with used in Chinese herb are observe the effects of Astragalus Danshen on rat isolated salivary glands. And after joining the Carbamoyl choline (CCh) the two kinds of distinct herb are on the influence of salivary gland secretion. The expression distribution of aquaporin-5 (AQP5) in submandibular salivary glands of SS on rat is shown by Western blot analysis. In addition, we found the influence of Astragalus Danshen compound in submandibular salivary glands of SS on rat.Results: After immunization 7W of SS rats, thus producing pathological changes similar to the salivary gland in human SS. It suggested that a large number of lymphocyte cells were different invasive depth and effective immunization. Through Chinese herb perfusion on rat isolated salivary glands, indicating that Danshen Astragalus could be promote salivary glands with secreted in different depth. Western blot analysis showed that there were a 42KD in each groups. The expressed numerus of the control group were normal(1.35±0.13),but the normal saline group were obvious abnorma(l1.03±0.08),There were clear enhancements of expression numerus on the Astragalus Danshen compound of high-dose and low-dose(1.31±0.15, 1.27±0.10). Compared with the control group, the expression of AQP5 in the normal saline group of rat model were a significant reduction (P<0.05); Astragalus Danshen compound of high-dose and low-dose were significantly increased the expression of AQP5 (P<0.05).Conclusion:1. Based on the Chinese herb on rat isolated salivary glands from perfusion, It discovered Danshen are slight to promote salivary glands directly before joining in no CCh. Similarly, Astragalus also have the same effect. Otherwise, the two distinct kinds of herbs are different to promote the secretion of saliva graphics.2. Astragalus Danshen compound through enhanced the release of water molecules in channel of submandibular glands, by floating the expression of AQP5, improving the secretory function and the local symptoms of salivary glands, increasing salivary secretory volume of SS rat model and reducing the damage degree of glandular organ, so as to release the xerostomia symptoms of SS.
