Objective:In order to make the medecine gathering at the corium tier, and make the treatment to the chronic eczeme maximumly, In this project,we make the spray of eczema cream,and contrast to the general spray, In this project, we studied the prescription sifting, preparation, quality standard and assess, percutaneous permeability from clinical prescription, and our research work provides scientific support to nanotechnology cream pharmaceutical of traditional Chinese medicine treating chronic eczema.Methods(1) Using hierarchy consistency examination, and use the clinical experiments as evaluating indexs, optimized the prescription for cream preparation.(2) Using High Performance Liquid Chromatography (HPLC) determination, built the determination method of oxymatrine and berberine hydrochloride.(3) Using single factor experiment, optimized the best surfactant and Oil phase,and researched the the best proportion parts of nanoemulsion through drawing pseudo-ternary phase diagrams, prepared the spray of eczeme nanoemulsion, using the mastersizer alginate-chitosan microcapsules measure particle size and distribution,using isothermal accelerated tests,studied the stability of spray of eczeme nanoemulsion; Detection of the package rate and drug loading by Sephadex column chromatography. (4) Using the content of matrine as evaluating indexs,studied the Storage capacity and absorpt speed of spray of eczeme nanoemulsion;Results(1) The prescription of the spary is:paeonolum B, Oxymatrine B, Oxymatrine C, borneol and mentho are both A, the grade sum R1 is maximun, and the curative effect is better than others.(2) The determination of Oxymatrine and berberine hydrochloride indicated that the linearity extent of standerd curve,the precision of instruments,the stability and reduplicate of sample solution,the recovery werecorresponded to the requirement of quantify analyse,which could be used for evaluating the process,quality and external absorbtion.(3) The best surfactant is RH40, and the best oil phase of nanoemulsion is IPM, when the quality contrastion of RH40 and IPM was 4:1, the nanoemulsion was steady,equal,diaphanous,and the colour is yellow. Matrine encapsulation efficiency was (87.06±0.5249)%. The equal particle size was between 23.1~115.8nm. The nanoemulsion issteady at-5~65℃and the relative humidity was 75%.(4) t50% of the spray of eczeme nanoemulsion was 1.782 times than the spray of eczeme, and the storage capacity of skin was 1.994 times as great as another. It indicated that the absorbing speed of the spray of eczeme nanoemulsion was slow, and the remain time was long, and the storage capacity of skin was high. ConclusionsThe prescription of the spary was reasonable, and the preparation of the nanoemulsion is reasonable,the quality was controllable, and the sample was steady. The skin storage capacity of the spray of eczeme nanoemulsion was larger than the spray of eczeme,and the absorbing speed is slower,so it correspond to the design request of skin,after clincal applyment,the curative effect of treating chronic eczema was even more remarkable.
