ObjectiveTo induce the apoptosis of osteosarcoma cell line MG-63 using cell mitosis inhibitor nocodazole as inducer and observe its biochemical features.MethodsFirstly, we studied the effect of gradient concentration of 1 nmol·L-110μmol·L-1 of nocodazole on MG-63 cells, then, we used 10,50,100,500nmol·L-1of four concentrationns of nocodazole to induce the apoptosis of MG-63 cells, The MTT colorimetric method was used to test MG-63 cell growth curve; Flow cytometry (FCM) analysis of cell cycle;Immunocytochemistry to detect the expression of PLK1; The expression of PLK1 of ost-eosarcoma cells line MG-63 was analysed by RT-PCR.ResultNocodazole role in MG-63 cells after 24h,Nocodazole significantly inhibited proliferation of colonies in MG-63cells, G0/G1 phase cells decreased, increase in the proportion of cells in S, G2/M phase cells was significantly increased,Compared with the control group was significantly(P<0.05).After nocodazole role, immunocytochemistry showed,PLK1 expression was significantly decreased. PLK1 mRNA expression of osteosarcoma cells line MG-63 declined obviously than before medication.ConclusionNocodazole induced apoptosis in MG-63 cells, G2/M phase arrest by inhibiting the mechanism may be implemented PLK1 gene expression.
