Objective:By studying the expression of follicle-stimulating hormone receptor (FSHR) gene and luteinizing hormone receptor (LHR) gene in granulosa cells in Polycystic Ovarian Rats' ovaris, under the influence of Bushen huayu decoction, the thesis discussed the clinical effects of Bushen huayu decoction and the connection between Genes of FSHR and LHR and Polycystic Ovarian Syndrome (PCOS), in order to explore the inner mechanistic about how Bushen huayu decoction cure PCOS from the molecular level, thus to provide the theoretical basis for clinical application.Method:79 SPF SD female rats aged 6 weeks were randomly divided into 6 groups:high, moderate and low dosage of Bushen huayu decoction groups, Diane-35 group, blank control group and model group. The rats were moulded with Letrozole through intragastric administration, once a day,21 days in total. Use (Real-time) RT-PCR (SYBRGreenl) technique to detect the expression of FSHR gene and LHR gene.Results:1. The expression of FSHR gene and LHR gene in granulosa cells in model group was obviously declined, compared with blank control group, there was significant difference (P<0.01) 2. The expressions of FSHR gene and LHR gene in high and low dosage of Bushen huayu decoction groups and Diane-35 group were increased remarkably, compared with model group, there was significant difference (P<0.01). However, no significant difference was showed between moderate dosage of Bushen huayu decoction group and model group (P>0.05)3. Compared with Diane-35 group, the expressions of FSHR gene and LHR gene in high and low dosage of Bushen huayu decoction groups had no significant difference (P>0.05)4. There was no significant difference in the expressions of FSHR gene and LHR gene between high and low dosage of Bushen huayu decoction groups (P>0.05)Conclusions:The research shows that the decline of expressions of FSHR gene and LHR gene is connected with the pathogenesis of PCOS. Besides, Bushen huayu decoction can increase the expression of FSHR gene and LHR gene in granulosa cells in Polycystic Ovarian Rats'ovaris, promote follicle to develop into dominant follicle, and make the ovulation successful, thus reach the therapeutic purpose.
