Prescription is composed by herbs which relates to compatibility, herb dosage, dosage form, sume methods and so on. Formula and the syndrome refers to particular prescription adapt or attending syndrome which usually involves pathomechanism and syndrome. In Chinese medicine system, "Relation among the herbs, formula, effects and syndrome" is not only the important academic logic of making a point but also experiential foundation of clinical treatment based on syndrome differentiation. According to the principal of "The correlation between formula and syndrome", effect of prescriptions not only have close relationship with prescription-formulation factors but also with the state or disease.In the light of the"Relation among the herbs, formula, effects and syndrome" the effect of tonifying qi and abating fever of Buzhong Yiqi Tang is to be reseached as the object. On the base of establishing the rat model of fever of spleen-qi deficiency, effect after using Buzhong Yiqi Tang and its different compatibilities have been observed.The compatibility rationality of Buzhong Yiqi Tang, and the relevance between Chinese medicine prescription and syndrome also dicussed.Research 1:To make fever of spleen-qi deficiency model in ratsMethods:the male Wistar rat;200-220g,40; The male rats were assigned into normal group, the pure fever group, the pure spleen-qi deficiency group, the fever of spleen-qi deficiency model group,10 rats in every group. the pure spleen-qi deficiency group and the fever of spleen-qi deficiency model group were placed in a large plastic bucket which filled with warm water (22±1℃) at 2:00 pm and swam 10 min, every other day feeding (every other day ban food, enough to eat every other day) for 3 weeks; normal control group did not exert any stimulating natural feeding. Every week we measured the body weight, temperature of rats and observed the change of Syndrome Model of autonomous behavior. All rates were feed with 10% D-xylose 5mland were placed in metabolism cage to collect urine of 5 hours. At 8:00 am on the 22th day, the pure fever group and the fever of spleen-qi deficiency model group were intraperitoneal injected with LPS (80(?)g/kg) to induce fever. Normal group and the pure spleen-qi deficiency group were intraperitoneal injected with physiological saline. The rectal temperature was measured at 30min, 60min,120min,210min,270min,330min after LPS injection and draw mean body temperature curve. It was followed by intraperitoneal injection 10% Chloral Hydrate (0.35ml/100g Weight) Draw blood 6ml from aorta abdominalis when the rats were narcosis.3ml was centrifuged 15min at 2500rpm, took the serum to measure GAS, IL-1β,IL-2,IL-6, T3, T4, TSH. another 3ml was placed in centrifuge tube which filled with 10%EDTA·Na230μL and Aprotinin 40μL centrifuged 10min at 2000rpm, took the plasma to measure MTL,SS.Result:1) Compared with the normal group, the rectal temperature of the pure fever group rat rises significantly, and at 60min,120min,180min,240min,330min has the more significance (P<0.05 or P<0.01); the rectal temperature of fever of spleen-qi deficiency model group has a transient decrease at30min, then raised obviously, and at 60min,120min,180min,240min the difference has the more significance (P<0.05 or P<0.01); the rectal temperature of pure spleen-qi deficiency has a transient increase at 60min with the significant difference (P<0.01),then has no change. Compared with pure fever group, the rectal temperature of fever of spleen-qi deficiency group has a lower level, the temperature curve is lower, and at 30min,60min,120min,330min the difference has the more significance (P<0.05 or P<0.01); the rectal temperature of pure spleen-qi deficiency group has decreased at all point-in-time except 60min, and its difference have significance (P<0.05 or P<0.01). Compared with pure spleen-qi deficiency group, the rectal temperature of fever of spleen-qi deficiency group has a transient decrease at30min, then raised obviously, and at 120min,180min,240min the difference has the more significance (P<0.05 or P<0.01);2) In experiment period, the body weight of normal group and pure fever group increased obviously. Compared with the normal group, the body weight of fever of spleen-qi deficiency group and pure pleen-qi deficiency group decreased obviously at the end of 1w,2w,3w,and the difference has significance (P<0.05 or P<0.01).the body weight of pure fever group has no change. Compared with the pure fever group, the body weight of fever of spleen-qi deficiency group and pure spleen-qi deficiency have decreased obviously at the end of 1w,2w,3w,and the difference has significance (P<0.01). Compared with the pure spleen-qi deficiency group, the body weight of fever of spleen-qi deficiency group has no significance (P>0.05).1 week after modeling, the normal group, the pure fever group, the pure spleen-qi deficiency group, the fever of spleen-qi deficiency model group all have all did not no change in behavioral activity, emotional reactions, skin hair, diet, sleep and defecate state.2 weeks after modeling, the pure spleen-qi deficiency group, the fever of spleen-qi deficiency model group both showed reduced activity, reduced the level of activity, food intake, furring and fair unsmooth, loose stool, etc.; 3 weeks after modeling,the model group began to slow down the movement, emotions ennui, food intake, fur and fair unsmooth, loose stools; etc. Compared with the normal group, the integral quantity of change in appearance behaviour were all increased, difference have the more significance (P<0.01); the integral quantity of change in appearance behaviour have no significance (P>0.05). Compared with the normal group and the pure fever group, pure spleen-qi deficiency group and the fever of spleen-qi deficiency model group are lessened in urine D-xylose excretion rate, peripheral blood GAS, MTL,IL-1β, IL-2,T3,T4,TSH increased in IL-6 and SS, those difference all have the more significance(P<0.05 or P0.05).Compared with the pure fever group, urine D-xylose excretion rate of pure spleen-qi deficiency group and the fever of spleen-qi deficiency model group have decreased, the difference all have significance (P<0.05).Compared with the pure spleen-qi deficiency group, peripheral blood T4 of the fever of spleen-qi deficiency model group have decreased obviously(P<0.01),other indexes have no significant(P>0.05).Conclusion:By adopting excess fatigue+out of constant diet+LPS injection had induced the rats show a serial of appearance in syndrome model of autonomous behavior, body weight, body temperature which were similar to Chinese symptoms of fever of spleen-qi deficiency. At the same time the model had pathological and physiological change about PB medium, gastrointestinal hormone, material metabolism hormone which were were similar to Chinese symptoms of spleen-qi deficiency. The rectal temperature was measured at 30min, 60min,120min,210min,270min,330min after LPS injection and draw mean body temperature curve. When the rectal temperature measured finished, the rats were decapitated to collect blood, 3ml was centrifuged 15min at 2500rpm, took the serum to measure GAS, IL-1β, IL-2, IL-6, T3, T4, TSH. another 3ml was placed in centrifuge tube which filled with 10%EDTA-Na230μL and Aprotinin 40μL centrifuged 10min at 2000rpm, took the plasma to measure MTL,SS. This indicates Buzhong Yiqi Tang has prevention and cure effect on the fever of spleen-qi deficiency model and its defervescence is connected with reducing central pyrexia amboceptor PGE2 and cAMP.Buzhong Yiqi Tang Qu Chaisheng group rats have no obvious improvement in appearance behavior, body weight has increased. blood GAS,MTL,T3,T4,TSH, IL-1βand urine D-xylose excretion rate has raised, SS has decreased; body temperature increased, Peripheral Blood IL-6 and hypothalamus PGE2,cAMP were all increased. This indicates Buzhong Yiqi Tang dedutced Chaihu, Shengma has partly effect on this model, mainly involved in the stomach and intestine absorption, metabolism, immunologic function,has no effect on fever.Buzhong Yiqi Tang Daji Chaisheng group rats have partly improvement in appearance behavior, body weight has increased. blood GAS, MTL, T3, T4, TSH, IL-1β,IL-2 and urine D-xylose excretion rate has raised,SS has decreased; body temperature increased clearly, d in the stomach and Peripheral Blood IL-6 decreased and hypothalamus PGE2,cAMP were all increased. This indicates increased Chaihu and Shengma dosage of Buzhong Yiqi Tang has partly effect on this modelmainly involve intestine absorption, metabolism, immunologic function, has no effect on fever.Daji Chaisheng group rats have no improvement in appearance behavior and body weight, blood GAS,MTL,T3,T4,TSH, IL-1β,Il1-2 and urine D-xylose excretion rate has decreased, SS has raised; body temperature decreased clearly, hypothalamus PGE2,cAMP were all decreased. This indicates large dosage Chaihu and Shengma have no effect on spleen-qi deficiency, have rominent effect on anti-fever.Conclusion:the integrated effectiveness of "YIQIJIANPI, SHENG YANG TUIRE"of Buzhong Yiqi Tang is closely related to selection of usage and dosage of Chaihu and Shengma, that is usage or large dosage compatibility of Chaihu, Shengma will influence the effectiveness of primary prescription. Compatibility of small dose of Chaihu, Shengma will make the best effectiveness of Buzhong Yiqi Tang.This topic has replicated Fever of spleen-qi deficiency rat model successfully. it not only enriches the content of Chinese syndrome model but also offers useful tools for experiment research of traditional Chinese medicine. Effectiveness comparation between Buzhong Yiqi Tang and its different compatibility offers experiment reason for proofing "Yiqi Tuire" function of Buzhong Yiqi Tang and experiential rationality on usage and small dosage compatibility of Chai hu, Shengma. In this research, different prescription compatibility takes different effect when the same syndrome of fever of spleen-qi deficiency, and this provide understanding from angle of modern pharmacology on connotation of "The correlation between formula and syndrome".
