Objective:To observe the effect of Run Fu Zhi Yang Cream (RFZYC) on the cell of human epidermal keratinocytes in vitro, and investigate the influence of Run Fu Zhi Yang Cream on P38 mitogen activated protein kinase(P38MAPK) pathway and vascular endothelial growth factor (VEGF) in keratinocytes. Research the relationship between P38MAPK pathway and VEGF expression.Methods:1. Normal epidermal keratinocytes isolated from human were cultured in the modified serum-free medium in vitro, and observe the change of cell morphology under inverted microscope. The cells'certain antigens were detected with immunohistochemical method.2. Keratinocytes were treated with different concentration of Run Fu Zhi Yang Cream, and test the without cell toxicity concentration range to the keratinocytes by MTT method.3. In non-toxic concentration range dilute four different concentrations of Run Fu Zhi Yang Cream, another plus tazarotene group, inhibitor group and control group. The cell proliferation activity was detected by MTT method.4.P-p38 expressions were assessed by western blot; VEGF in the supernant liquid was measured by ELISA.Results: 1. Cultured cells in vitro had the normal form of keratinocytes and were stained positive for keratin.2. Run Fu Zhi Yang Cream had no cell toxicity to keratinocytes during 0-0.4855g/ml concentration range; Compared with the control group, Run Fu Zhi Yang Cream could significantly inhibit the proliferation in the concentration of 0.4 g/ml,0.2 g/ml and 0.1 g/ml (P<0.01), and in the concentration of 0.05g/ml it could promote the proliferation, but the difference was not statistically significant (P> 0.05). Compared with tazarotene group, in the concentration of 0.4g/ml the difference was not statistically significant (P>0.05).3. Compared with negative control group, in the concentration of 0.4g/ml and 0.2 g/ml, P-p38 protein expression reduced significantly (P< 0.01); In the concentration of 0.1 g/ml, P-p38 protein expression reduced (P< 0.05). Compared with negative control group, the P-p38 protein expressions of tazarotene group and inhibitor group reduced significantly (P<0.01).4. Compared with blank control group, the VEGF secretion was significantly declined in the concentration of 0.4g/ml and 0.2g/ml (P< 0.01); In the concentration of 0.1 g/ml, the VEGF secretion was declined (P<0.05); Compared with blank group, the VEGF secretion had no obviously change after the keratinocytes treated by inhibitor (P> 0.05). Conclusion:1.The Run Fu Zhi Yang Cream can inhibit the proliferation of keratinocytes in a certain concentration range, and the function of it has no difference with that of tazarotene.2. The Run Fu Zhi Yang Cream can cure psoriasis, and the mechanism of it maybe due to the activation of P38MAPK signal pathway, thus plays inhibition effect of keratinocytes proliferation.3. The Run Fu Zhi Yang Cream can reduce the VEGF secretion, however, it has no relationship with the activation of P38MAPK signal pathway.
