Objective:To investigate the feasibility that transplantation of human umbilical cord mesenchymal stem cells (HUCMSCs) in the treatment of diabetic rabbit foot of deficiency of Yang and stasis of blood, aim to provide a more manifold, more security, more effective method to cure diabetic foot of deficiency of Yang and stasis of blood in theory.Material and method:①Models of diabetic rabbits were established, give alloxan by vein of auris edge in 30 seconds, monitoring glucose, avoid hypoglycemia, Monitoring Glucose that 48,72hours, preponderate over 11.1 milimore per litre,②Model succeed after 72 hours, the general anaesthesia is chloramine-ketone is combined with haloperidol injection. From groins ligament to knee joint, we make out a vertical incision to separate and ligate femoral arteries and ramus.③Prepared HUCMSCs in sign of DIL were directly injected into the left hindlimb adductor and gastrocnemius muscle in diabetic rabbit models (cell quantity was 1.67×10~6 per point). Saline was directly injected into right lower limbs.④At 15 and 30 days, adductor and gastrocnemius muscle of the bilateral hindlimbs were collected. Pathological sections was observed. Capillary density and skin temperature were measured using immunohistochemical staining for CD31.Results:1. The skin temperature was significantly greater in the transplantation group compared with control group (P<0.05).2. Pathological sections exhibited that collateral vessels were significantly increased in the transplantation group compared with control group.3. Immunohistochemical staining for CD31 demonstrated that the capillary density was significantly higher in the transplantation group compared with control group(P<0.01).4. After HUMSCs transplantation in 15 and 30 days, in angiography, transplantation group,s capillary vessels are make up obviously in 15 days, in contrast, the control group only expand the vessels and the capillary vessels are not make up obviously; the transplantation group,s capillary vessels get thicker in 30 days than 15 days. There are capillary vessels more less treatment group.Conclusion:HUCMSCs transplantation in the treatment of diabetic rabbit foot of deficiency of Yang and stasis of blood can improve the lower limbs temperature, establish the lower limbs more side branch and improve the lower limbs capillary more vessel density. Transplantation of HUCMSCs is a safety, effective and therapy for diabetic rabbit foot of deficiency of Yang and stasis of blood.
