The extraction of Cephalospori11 C (CPC) from the whole brot11 byaqueous two-phase system is discussed. The effects of parameters suchas PEG lnolecular weight, PEG co11ce11tration, the type al1d theconcentration of inorganic salt, plI, temperature and the concentr8tionof Cepllalospori11 C ol1 tl1e Cephalosporin C and DesacetylCephalosporin C partition coefficiellt are studied. The optimum aqueoustwo-phase system is 3l% PEG400 / 24% (Mb)2SO4, (Kc=5.9l, tl1erecovery yield=90.l%). For chal1gi11g tl1e partition coefficient, theaddition of solvents and neutral i1lorganiG salts is also investigated. Tl1eextraction of Cepl1alosporin C fTom whole broth, counter-eXtraction,multistage extraction and tl1e crystallization by iso-propylalcohol is used. The whole recovery yie1d is 52.7%. |
