Hcpatic fibrosis rcfcrs to thc pathologic proccss which is characterized by the exceeding amount of fibrous tissue in liver. This prucess would lead to cirrhosis in many chronic liver dc~es. The modern conception of hepatic fibrosis is summarized as the result of more synthesis and less degradation of extra-cellular matrix(ECM). There~ia high incidenc.c~ of hepatitis 1~ in China; 1l~ 15 ShOWn lfl SOfl~- reports that the majority of cirrhosis result from chronic viral hepatitis, and tTht( cirrhosis is 潼 irreVersible disturbance of hepatic ar駅ct黵e. So the focus of research is to effectively control or reverse the phase of hepatic fibrosis. The recent therapeutic study on hepatic fibrosis was pnnuririly concentrated on metabolism of collagen and other ECM to seek drugs which cain inhibit collagen synthesis or precipiLik collagen degradation. But their effects were not satisfactory. In addition, because of a variety of adverse effects, the applications of these drugs are limited to the great e~xtent. 1CM had been showed to have particular advantages in the therapy of hepatic fibrosis. In the tight of its pathogenesis in TCM that include blood stasis and body invigorousness, we composite a 1CM prescription, (3ANXIANWAN(GXW), according to the principle of enhancing Qi抯 function and promoting blood circulation. It had been tested by years of the clinical practice of Professor. Yu Shaoy win, m9 (黷or, that the prescription are welt suit for patients with hepatic fibrosis. And its effect is satisfied and reliable. To study the mechanism of Anti-fibroses effects of the compound mentioned above~ we.- had devc.lop both c.tinic.al and experimental researches in which GXW was administered to the treatment group with Da~ui,~2heChongWatn(DHZ) used as control. The results in animal experiments showed GXW reductions of such rats?serum marks as HA, LN, P11W, and collage IV in treatment group, which indicated anti-fibrosis effects of the compound, in addition, it can also improve liver function, that is, the levels of the serum. ALT, Cub and Alb in eatfttent group were lower than that in con鼅ol group. The biopsies suggested that there were less severe liver injury in the treatment group than that in control. Under light microscopy; less necrosis or degeneration of hepatocytes and- proliferation of hepatk- 3 fibrous tissue were observedin rats treated with GXM. The observation of oi~anelles under electron microscopy also showed the similar improvement in treatment group. According to statistical data, the total efficacy of the TCM compound, especially with the administration ot higher dosage, was superior to that of DHZ. In the clinical investigation. GXW effectively relieved sympkmns with hepatic fibrosis and protect liver function by reducing serum level& of ALT, Alb and elevating Glb level. Above all it can degrade ECM, the level of whose serum marks such as HA and PCIII all decreased significantly. In addition to these effec is, the compound also contribu(e to the HBe seroconvemion in patients treated with the TCM compound. In conclusions (JXW have obvious therapeutic effect on hepatic fibrosis which results from chronic Hepatitis B The fimc.tioti Protecting hepatocytes and Anti4ibrosis effect should contribute to its line efficacy.
