Abstract Aim To study the effect and mechanism of ischemic Preconditioning and sodium aescinatein in the acute ischemia-reperfusion model of rat brain by 4 vessels block. Methods Base on closed double carotid vessel by artery nip 4 simultaneity after 1 day the vertebra artery of rats,the animals were cauterized and closed . The rats divided into 5 groups. 1) model group ,30 minute global ischemia and reperfusion 30 minute; 2) preconditioning group 30 minute global ischemia and reperfusion 30 minute after repeated 3 times 3 minute ischamia and reperfusion 3 minute;3) model treated sodium aescinate group ,treated 10 mg/kg ; 4) combine preconditioning and treated sodium aescinate group ; 5)control group, exposed double carotid vessel and not closed. We observe the changes of scavenging oxygen tree radicals metabolim ,adenosine metabolism and the water quatities in the cortex and the serum. Results Compared with control group ,model group showed a significant increase in concentrations LPO,NO ,but a significant decrease activities SOD,GSH-Px ,CK-mb and contents of ATP,ADP,AMP in the cortex and the serum.lt also showed a increase water quantities of the cortex. The all above data in preconditioning group and model treated sodium aescinate group are improved obviously .lt showed significant improved in combine preconditioning and treated sodium aescinate group. Conclusion 1 .The acute ischemia- reperfusion model of rat brain by 4 vessels block, can be used for the study of ischemia preconditioning in the brain. 2.The protective effects of preconditioning and sodium aescinate maybe related with scavenging oxygen free radicals, decreasing NO and producing adenosine .3.Combine preconditioning and treated sodium aescinate group is the best way in the brain protection.
