0BJECTIyE: To investigate the effect and mechanism ofPo1ygonatum Mill po1ysaccharide (PM) eyedrops on experimenta1 dryeye.wt: In 23 Janpanese white rabbits (46 eyes), 18 rabbits (36eyes) were induced into dry eye mode1s by escion partia1 1acrima1 g1andand Harder's gland and a11 the third 1id, whi1e 5 rabbits (10 eyes)were norma1 group. Ten days later 30. dty eye mode1s were se1ected andrandomized into three groups: observation group (5 rabbits, 10 eyes),positive contro1 gToup (5 rabbits, 10 eyes) and negative contro1 group(5 rabbits, 10 eyes). The three groups,ere dea1ed 'ith solvent, 0. axPM eyedrops and Tear Natural (TN) in bilatera1 eyes respectiveiy.Schirmer 1 test (S1t) and Rose bengal staining (Rb) of cornea andconjunctiva were done once a week. 5,eeks 1ater the tissue morphologyof 2 cornea chosen randomly from each group were examined undertransfer e1ectronic microscope. The tissue morphology of 2 1acrima1g1ands and Harder's g1ands chosen randomly from each group andconjunctiva of a11 rabbits were examined under optica1 microscope.Conjunctival gob1et--ce1ls were counted. In clinica1 l5 patients (30eyes) with dry eye had been treated with 0. 8% PM eyedrops for 5 weeks.The subjective dry eye spoptom, Slt, Rb and tear Break--up time (BUT)were measured pre-- and posttreatment.REAULTS: (1) In norcal rabbits the mean 1ength of Slt is 13. 45l2. 4lmm and the mean conjunctival goblet--ce11 density is 22. 040l3. 0995 per microscopic field of 400 times. (2) 8efore treatment thesynthetica1 rank of S1t and Rb were similar in the three groups, andno difference was detected (P>0. 05). (3) The Slt and Rb change amongthe three groups showed signifecent difference at 2 and 3 weeks aftertr6atment respective1y (P<0. 05). In observation group 2 and 3 weeksrespectively after the treatment S1t began to better remarkablycompared with those in negative group (P<0. 05). 4 weeks after thetreatment the Rb of positive control group was better than that ofnegative nontro1 group. (4) In observation group plenty of microvil1iand microplicae were examined. There is a statistica11y significantdifference in gob1et--cel1 count between observation group and the twocontro1 groups (P<0. 05). A difference between normal group andobservation group was noted (P<0. 05).C0NCLUI0N: The use of 0. 8% PM eyedrops can resu1t insignificant1y more recovery of the ocu1ar surface of dry eye mode1sand inhibit efficien1y dry eye.
