ApoLipoprotein E (Apo E) gene which is closely linked with Apo C I and Apo C LI genes on chromosome 19 queue, is genetically polymorphic at two Ioci encoding the amino acid residues 112 and 158. We have used restriction isotyping (restriction enzyme isoform genotyping) for rapid typing of common apolipoprotein E isoforms (E2~ E3E4). In the present study, a 244bp DNA fragment in the fourth exon of Apo E gene was amplified by polymerase chain reaction(PCR),and subsequently analyzed by digestion with the restriction enzyme Hha I, followed by 4% low melting point agarose gel or 20% nondenaturation polyacrylamide gel electrophoresis of the cleavage products. The Apo E genotype was determined in 102 healthy ChinaKorean individuals who had been in Yanbian at least 3 generation. Apo E allelic frequencies were , 0.083 for E2 , 0.833 for E3, , and 0.078 for E4, respectively. Compared with Chinese population and other nationalities, there was no difference of allele frequency. Geographic and ethnic comparisons had shown that E4 allele was more common among Caucasian and less common among most Asian.
