Objective To isolate feta~ nucleated red blood cells (}7NRBCs) from maternal blood ,which be used in noninvasive prenatal genetic diagnosis Methods Peripheral blood mononuclear cells (MNCs) were seqarated by single density gradient Histopaque-1077 from 32 pregnat women with gestations behve~n 1 b2C) weeks ,FNRJ3Cs were entiched from the M7NCs by flow cytometiy using monoclonal antibody against the glvcophotin A(GPA).Sortecl cells of male fetuses in maternal blood were con県med usim i-special DNA sequences (SRi) by polytnerase chain reaction ( PCR)and by Y梒liiomosotne fluorescent staining. Results (193 ? i0~?. ii X I (Y3GPA positive cells per milliliter in pregnant women pcaipheral blood were softe(l by the flow cytornetry.More 90% results of PCR were identical with the g~n~kr dkr ahorticide.
