Objective:To investigate the regulating action and its mechanism of Bushenzhuanggu Capsule(BC) on bone resorption-formation discoupling in Diabetic Osteoporosis(DO) rats models induced by Alloxan(ALX) intraperitoneally. Methods:The rats were randomly divided into five groups,ie. model control,diabetes osteoporosis treated with low dowsage of BC,middle dosage of BC.high dosage of BC,and Calcium Carbonate & Alpha D3 respectively. Normal control group is also established. The effect of different medicines on the following parameters were detected by radioimmunoassay,immunohistochemistry and histomorphometry,etc. which were serum level of insulin,C-peptide,PTH,CT,25-(OH)VD3,expression of transforming growth factor- P 1 and caspase-3 in rat osteoblasts,histomorphmetric analysis of bone structure,changes in general condition and blood glucose as well. Results:The abnormality of parameters stated above were shown in DO rats.While BC demonstrates the action of (1) improving the general condition,regulating the metabolism of blood glucose and correcting insulin deficiency partially.(2)modulating the metabolic disorder of calcium-regulating hormones such as PTH CT VD3 (3) promoting the expression of TGF- B1 in osteoblasts to regulate local growth factor level in bone.(4)stimulating the recruitment and/or proliferation of osteoblasts as a result of inhibiting the expression of caspase-3 in osteoblasts .(5) increasing the bone mineral contents and improving the microstructure of bone. The comprehensive curative effect of BC is superior to that of Calcium Carbonate combined with Alpha DS. Conclusion:BC acts on bone resorption-formation coupling through many pathways and links such as the level of whole body,local organ,cell and molecule,etc. which contributes to the prevention and treatment of Diabetic Osteoporosis.
