This paper reviewed the pathbiology mechanism of Alzheimer Disease and the advance of the nootropic drugs .Based on it , the writer took the piracetam which has displayed the excellent ability of improving cerebral insufficiency as the leading compound . Starting from pyrrolidinone, eighteen target compounds were designed and synthesized ( N,N'-dihydrazide compounds ) to find new nootropic drugs , seventeen of them had not been reported in literature. The structures were identified by physic-chemical properties and spectral analyses ('H-NMR, IR ).The step-down tests in mice of fourteen compounds had been taken by two dosages. The result showed that HYCL-1, HYCL-3, HYCL-5, HYCL-6, HYCL-12, HYCL-14, HYCL-15 had good nootropic activity (there is significant difference in the number of errors and the escaping latency) , while HYCL-8, HYCL-11, HYCL-16 had mild or no activities (there is no significant difference in the number of errors and the escaping latency) . None of them had stronger nootropic activity than piracetam.
