ObjectiveTo observe the effect of Gancaosanshen granule on transmembrane potential(TMP) during rat s myocardial Ischemia-Reperfusion leading to cardiac arrhythmia. Meanwhile this research try to investigate the effect of Gancaosanshen granule on Nitric Oxide production in blood of right atrium and the activities of NOS and Na+-K+ATPase in myocardium of left ventricle. It shows the role of Nitric Oxide and Na+-K+ATPase during the functional mechanisms of Gancaosanshen granule. To study and reseach the influence and principle of arrhythmia by Gancaosanshen granule not only provide scientific basic for clinical treatment but also give a reference to develop many efficicent and specific antiarrhymia' s drugs. MethodsWe randomly divided the forty rats into five groups. Every group had eight rats. The experiments were performed as follows: A.normal group. B.sham group (rats without ligation of left anterior desending artery). C.control group (rats with ligation of left descending artery for 10 min followed by 30 min reperfusion).D.gancaosanshen granule prevention group(build model after rats treated with gancaosanshen granule for seven days).E. Propafenone contrast group(build model after rats treated with Propafenone for seven days). We detected the transmembrane action potential by floating microeletrode. We recorded the electrocardiogram in standard lead II of ECGA11 information was put into RM-6280.The change of the blood levels of Nitric Oxide in right atrium, the activity of myocardial Nitric Oxide Synthase and the Na+-K+ATPase activity were observed. Results1 Compared with normal group and sham group, ECG and TMP of control group were changed significantly(P<0.01). The incidence of arrhythmia rats is manifestly lowered after rats were perfused gancaosanshen granule(P<0.01).At the same time TMP of gancaosanshen granule prevention group were apparently ameliorated.lt was not obvious between gancaosanshen granule pervention group and propafenone contrast group(P>0.05).2 The activity of Na+-K+ATPase in control group was significantly less than normal group(P<0.01). Gancaosanshen granule increased the Na+-K+ATPase activity(P<0.01). However, it was not a apparent diffierence between gancaosanshen granule group and propafenone group.3 Myocardial constitutive NOS(cNOS) activity and total NOS activity decreased significantly in control group(PO.01).NO production was reduced during I/R in control group(P<0.01).NO level during I/R increased significantly(P<0.01),and myocardial cNOS (PO.01) and total NOS(PO.Ol) 30 min after reperfusion maintained higher activities in gancaosanshen granule group. These were compared with control group.The level of NO between gancaosanshen granule group and propafenone group was not a obvious discrepancy(P>0.05).ConclusionThe result indicated that the gancaosanshen granule can redress the electrocardiogram and ameliorate transmembrane action potential of the cardiac arrhythmia rats. Meanwhile the gancaosanshen granule can improve the blood level of NO in right atrium and the activities of NOS,cNOS and Na+-K+ATPase. All show the gancaosanshen granule prevents the cardiac arrhythmia.
