In Addition To The Mouse Cardiac Screening And Cloning Of Target Genes Regulated By Hsf1 In Heat Shock Response, Hsf1 Gene Knock

OBJECTIVE: To explore and clone genes regulated by heat shock transcription factor 1(HSF1) in the myocardium during heat shock response. METHODS: HSF1 knockout mice were subjected to heat shock treatment (42℃ for 15min, recover for 3h), and cDNA microarray containing 15000 mouse cDNAs were employed to investigate the expression of genes regulated by HSF1 in the myocardium. The promoter regions of differentially expressed genes were analysed by TESS software to search for heat shock element(HSE). Then down-regulated genes containing HSE in their promoter regions in the myocardium of HSF1 knockout mice were further analysed by RT-PCR. Furthermore, new genes differentially expressed were cloned by bioinformatics methods. RESULTS: 398 genes were down-regulated in the myocardium of HSF1mice during heat shock response, with 173 known. And 641 genes were up-regulated in the myocardium of HSF1 mice during heat shock response, with 235 known. Then, five of the down - regulated genes and five of the up-regulated ones were found containing HSE in their promoter regions. Furthermore , human XLHSRF1 and rat XLHSRF2 genes were cloned and submitted to GenBank. CONCLUSION: During heat shock response, the expression of multiple genes is drastically changed in the myocardium of HSFT mice. Ten ofthem were found containing HSE in their promoter regions. It was indicated that these genes may be directly regulated by HSF1. Two stress-related new genes were cloned with ioinformatics methods. This study laid foundation for further investigation of the functions and roles of these genes.
