Aspirin, Tetramethylpyrazine Phosphate Compatibility Antithrombotic Effect And Its Mechanism Is Discussed

The antithrombolic effects of Asp/Lze and its mechanism involved were investigated.The results showed that Lze sum Asp(1:1) could produce a synergistic effect.The single-dose administration of Asp/Lze (ig 100/100mg/kg) apparently prolonged cruor time in vitro and the bleeding time of tail in mice .Asp/Lze(ig 50/50,100/1 00mg/kg) could remarkably decreased the pulmonary thrombus mortality of the mice induced by collagen-adrenaline .Asp/Lze(ig 35/35,70/70mg/kg)significantly decreased the experimental thrombus length ,wet weight and dry weight in rats .In addition ,Asp/Lze (ig 18.5/18.5,37/37mg/kg)could not only refain the formation of carotid artery thrombus, but also dissolve carotid artery thrombus by cotton thread in rabbits. Fibin degradation products(FDP) level was significantly increased.Platelet aggregation induced by thrombin,collagen and ADP were all markedly inhibited by ig Asp/Lze in mice(50/50,100/1 00mg/kg), rats(35/35,70/70mg/kg) and rabbits (18.5/18.5,37/37mg/kg) .The single-dose administration of Asp/Lze(ig 35/35,70/70mg/kg) could apparently inhibited platelet adhesion in rats .At the same doses for 7 successive days, Asp/Lze could remarkably decrease blood viscosity and shortened RBC electrphoresis time ,increase the 6-Keto-PGF1a level and decrease the TXB2 level of plasmasample .The results of safety trial showed that Asp/Lze ig, LD50 was 2143mg/kg in mice . Meanwhile , it could not change the number of mouse autonomic activity and cat respiration or heart rate number and blood pressure .Summarizing all experimental results, antithrombolic effect of Asp/Lzemight involve that it significantly inhibited platelet adhesion, aggregation andrelease reaction .In addition ,it could improve the blood rheology . Dissolvion thrombus of Asp/Lze might involve that it could increase FDP level and depolymerize aggregated platelet.
