Gastric carcinoma is formed during the role of external factors { physical, chemical and biological Jactors) and internal factors. With the development of medicine research, biological carcinogen factors are thought more by people. In recent years, epidemiological studies have showed the infection of Helicobacter Pylori (Hp) played an important role in the histogenesis of gastric carcinoma. In 1994, Hp has been named as first carcinogen of gastric cancer by World Health Organization (WHO).It is proved another biological carcinogen factor - - the Epstein - Barr virus (EBV ) has close relationship wifh many kinds of carcinomas , recently people pay more attention to the relationship between EBV and gastric cancer. The incidence of gastric cancer which is positive of EBV infection is 18% in German, 16% in America,6% in Ja-rpan. In our coutry, the incidence of gastric cancer which is positive of EBV infection is 2 ~ 17. 5% , and 10% on average. In EBV - related tumors ,the incidence of gastric cancer is highest. .EBV latent membrane protein is a integrated membrane protein moleculer which plays important role during EBV transforming cells. It's the only protein which can transform epithelial cells coded by EBVand is regarded as EBV's oncogene. In our country ,there is high in-cidence of gastric cancer and the infection rate of EBV and Hp ishigh. It's reported by foreign research that 90% EBV - related gastriccancer has IM near the cancer tissue; on the other hand, Hp infection has been proved had close relationship with IM. Then if there is EBV infection in IM mucosa, and if EBV infection has coorperation with Hp infection during the development of IM and gastric cancer ? But there is no report about EBV -LMP's expression in IM mucosa and it's relationship with Hp infection by far.The present research aims at:①How is the EBV - LMP's expression in normal mucosa, in IM mucosa - the precarcerous lesion and in gastric cancer tissue. ②If there is coorperation between EBV and Hp during the development from IM to gastric cancer, further discussing the many - steps,many -factor's pathogenesis of gastric carcinoma.METHODThe biopsy samples of the study were all collected in gastroscopic screening from zhuanghe city, the high - risk area of gastric cancer. There were 20 cases of normal gastric mucosa, 171 cases of IM gastric mucosa,30 cases of gastric cancer which including 6 cases of high differentiated gastric adenocacinoma, 8 cases of middle differentiated gastric adenocacinoma, 12 cases of low differentiated gastric adenocacinoma, every 2 cases of myxedema and signet ring cell.Hp infection was diagnosed by histopathological examination he-matoxylin - eosin ( HE ) ; Hp infecton was confirmed or excluded by three tests: histologcal examination HE, Hp - DNA PCR and enzyme -linked immunosorbent assay ( ELISA) for anti - Hp immunoglobulin G (IgG) ;the subtypes of IM were classified by high - iron diamine/al-cian blue pH2. 5/periodic acid - schiff ( HID - Ab pH2. 5 - PAS) method; The single - cloned antibody of EBV - LMP were determinedby S - P immuno histochemical method.RESULT1. Expression of EBV - LMP in normal gastric mucosa, IM gastric mucosa and gastric cancer;There was no EBV - LMP expression in 20 cases of normal gastric mucosa(0% ) while the positive rates of EBV - LMP expression in 171 cases of IM mucosa(33. 3% ) were significantly higher than that of normal gastric mucosa( p < 0.05). The positive rates of EBV - LMP expression of gastric cancer(6. 7% ) were higher than that of normal gastric mucosa( p < 0. 1) and were lower than that of IM ( p < 0.05). The stain of EBV ?LMP was mainly in nuleus of cells.2. EBV - LMP expression in different subtype of IM mucosa: The study classified the subtypes of IM by HID - AbpH 2. 5 -PAS method. In 171 cases of IM mucosa, there were 54 cases of I subtype IM,76 cases of II subtype IM,41 cases of III subtype IM.
