Anti - Hepatitis Of Yinchenhao Development Of Effective Substance And Its Injection

Yinchenhao Decoction, a classical prescription in Shang Han Lun, consists of Herba Artemisiae Capillaris, Fructus Gardeniae andRadicx et Rhizoma Rhei. Being bitter and cold for inducing diuresis, clearing heat and dampness , normalizing functions of gallbladder and curing jaundice, reciding jaundice, it is an important herbal drug for treat jaundice. Some injections rooted from Yinchenhao Decoction have played important roles in the treatment of urgent, serious and infective hepatitis for a long time , but can't keep up with the development of modern traditional Chinese drug(TCD).So it is imperative to develop new injections from the prescription.After analyzed and optimized the prescription, this study exploited a new injection , Yinchen injection, whose prescription was made up of effective constitutes extracted from Artemisiae Cappillaris. At the same time, the preparation, stability, quality and fingerprint were investigated synthetically in this paper.By comparing comprehensively the water extract with ethanol extract and SFE CO2.extract, the water extract technique was selected and optimized according to the content of effective constitutes and extract yield for its economy and convenience. Ethanol was added into the concentrated waterextract to get rid of protein, starch, polysaccharose and so on. Then effective ingredients were enriched by ethyl acetate extraction and thetransfer ratio was more than 60%.This study built methods and standards for the quality control of medicinal material, stuff drug and Yinchen injection by HPLC and UV. The total content of scoparone, general organic acids and general flavones was more than 80%, including scoparone≥15%, general organic acids ≥10%and general flavones≥55%Injection water added solubilizers, PEG 400, could increase the solubility and stability of injection. Speeding up experiment and remaining sample experiment all indicated that Yinchen injection had perfect clarity and stability, which were both key issues for modern injection of TCD.For the first time, the fingerprints of the medicinal materials, stuff drug and Yinchen injection were set up. For sixty minutes, more than ten peaks could be separated on baseline in HPLC chromatogram. The correlations of fingerprints among medicinal materials, stuff drug and Yinchen injection were expounded by cluster analysis.Pharmacological experiment showed that Yinchen injection could obviously promote the bile excreting of rat model that was caused by ANIT, and almost had the same effect compared with the Yinchen compound injection, which had been used in clinical in 302 Hospital of PLA for more than thirty years.All above-mentioned foreclinical studies showed that it is worthy of further development of Yinchen injection.
